Partâ… Expression of Interleukin-18 and Interleukin-18 receptor in the Retina of Spontaneous Diabetic RatsObjective:To explore the location and expression of IL-18 and IL-18 receptor in the retina of different duration of type 2 diabetes rat model and to investigate the potential role of theirs in diabetic retinopathy.Methods:Select fifteen female spontaneous diabetic GK rats in the experimental group and fifteen male age-matched normal Wistar rats in control group(NC group). According to duration of diabetes GK rats were divided into three groups, that diabetes Mellitus group of 8 weeks(DM8 group),16 weeks(DM16 group),24 weeks(DM24 group), each group 5.The expression of IL-18 and IL-18R protein in the retinal tissue were detected by Western-blot. IL-18 and IL-18R mRNA expression in the retinal tissue were analyzed by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) technique. And the localization of IL-18 and IL-18R in the retinal tissue of type 2 diabetic rats were detected by immunohistochemistry.Results:1. Compared with the control group, the levels of IL-18 and IL-18R protein were higher in DM8 group (P<0.01),while weaker in DM16 group(P<0.05) and much weaker in DM24 group(P<0.01).Their differences were statistically significant.2. The expression of IL-18 mRNA were elevated in DM8 group than those in control group, while weaker in DM16 group and the lowest in DM24 group. The level IL-18R mRNA rised in DM8 group than those in the control group.But its expression in DM16 group were still stronger than its in the control group,till to decline in DM24 group.3. Immunohistochemistry showed that rat retinal tissue expression of IL-18 lied in the cytoplasm of ganglion cells, inner nuclear layer, outer nuclear layer and nerve fiber layer. But IL-18R staind in the cytoplasm of ganglion cells, nerve fiber layer and inner and outer plexiform layer axon.Conclusion:1. The expression of IL-18 and IL-18R involve in the development retinopathy of Type 2 dibetes.IL-18 plays an important role in the early stage of diabetic retinopathy.2. Changes in the expression of IL-18 may be involved in the occurrence of diabetic neural retina lesions. Retinal lesions in type 2 diabetes can occur early,and retinal degeneration of nerve tissue may be abnormal early in the vascular tissue.3. The role of IL-18 is not completely finished by IL-18R pathway, IL-18 may have other signal channels.Part II Expression of Interleukin-18 in the Retina of Diabetic Rat Models Induced by Alloxan MonohydrateObjective:To explore the location and expression of IL-18 in the retina of different duration of type 1 diabetic rat models induced by Alloxan Monohydrate and to investigate the potential role of theirs in type 1 diabetic retinopathy.Methods:Sixty female SD rats,weighted 220-250g,were randomly allocated to experimental groups(forty) and control groups (twenty).The experimental groups were intraperitionally injected with alloxan 200mg/kg to establish type 1 diabetia rat models. According to duration of diabetic rats were divided into three groups, that diabetes Mellitus group of 4 weeks(DM4 group),8 weeks(DM8 group),16 weeks(DM16 group), each group 8,8,6 respectively.The expression of IL-18 mRNA in the retinal tissue were analyzed by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) technique. And the localization of IL-18 in the retinal tissue of type 1 diabetic rats were detected by immunohistochemistry.Results:1. Compared with the control group, the levels of IL-18 mRNA were higher in DM4 group,while weaker in DM8 groupand much weaker in DM16 group.2.
