Objective:To investigate the relationship between expression of CD24,CD34 protein and carcinogenesis,development,the grade,clinical stage ,depth of invasion,lymphnode metastasis of gastric carcinoma. CD24,CD34 expression of 56 gastric carcinomas were detected. provide a theoretical basis in the early diagnosis,treatment and prognosis prediction for gastric carcinoma.Methods:CD24 protein expression of 56 gastric carcinomas were analysed by SP immunohistochemistry.The expression of CD24 protein was shown by the percentage of positive cells. the expression of CD34 was shown by MVD. The whole group of 56 cases were followed up。Results:1. CD24 expression in human gastric carcinoma: CD24 staining in the cytoplasm and membrane,In 56 cases of gastric carcinoma, 45 patients(80.36%) with CD24-positive。CD34 staining in vascular endothelial cell membrane. Microvascular staining was brown, and it showed heterogeneity distribution, the most intensive staining region "hot spots", mainly distribute in tumors edge and Cancer cells intensive areas,In all 56 cases of gastric carcinoma , MVD was 44.00±11.08。2.The relationship between CD24,CD34 and the clinical characteristics of gastric carcinoma: In 8 cases of early gastric carcinoma, 3 cases were weakly positive expression of CD24, in 48 patients with advanced gastric carcinoma, 38 cases were strongly positive or moderately positive, The difference was significant. (P <0.0001). Comparison of CD24 expression and the histological type of gastric cancer, there was no significant correlation(r=0.1901,P=0.1604). TNM staging and high expression of CD24 protein was significant, Comparison of TNM staging ofⅡ,Ⅲ, andⅣwithⅠ, the difference was significant (P values were 0.0010,0.0009,0.0003). Comparing the degree of gastric cancer cell infiltration in T2, T3, T4 with T1, the difference was significant (P values were 0.0047,0.0008,0.0005).In the lymph node metastasis, Comparison of N1, N2, N3 with N0, the difference is statistically significant (P values were 0.0027,0.0004, <0.0001). comparing MVD with the histological grade and lymph node metastasis ,there were no correlation (P> 0.05),and concerned with the depth of tumor invasion and clinical TNM stage (P <0.05).3. the relationship between CD24 expression with MVDand prognosis : the MVD'mean(×400) was 44.00±11.08. With the rate of CD24 positive cells increased, MVD has also increased, there were positively correlated (r = 0.72, P <0.0001). This shows that CD24 protein expression may be related to promoting the growth of tumor blood vessels. the higher the expression of CD24 protein, mean survival time was lower, and the differences between groups are statistical significance.Conclusions:1. CD24 protein expression in gastric carcinoma with the depth of cell invasion, the number of lymph node metastasis and TNM stage were positively correlated, this suggesting that CD24 protein plays an important role in the tumor occurrence and development.2. Comparing MVD with the histological grade and lymph node metastasis ,there were no correlation,and concerned with the depth of tumor invasion and clinical TNM stage.3.Comparing CD24 protein expression with angiogenesis in gastric carcinoma and cell proliferation ,there was positively correlated, The median survival time of The patients with CD24 protein higher expression was significantly reduced,this suggests that CD24 can be used as a symbol of tumor invasion and prognosis indicators.
