Aim: To study the effects of TARCD on the learning and memory of dementia model rats and its mechanisms. Methods: MG-2 type Y-electronic maze and morris water maze were used to study the behaviors of senile model rats and dementia model rats induced by scopolamine and D-galactose. To study the cell protective effects, the PC-12 cells were cultured and viabilities were measured with MTT method. In order to investigate the mechanisms of TARCD, the activities of several enzymes, the contents of protein and nucleic acid were determined. Microstructure and ultrastructure of rats' brain were also observed, and RT-PCR and immunohistochemistry were used to study the expression of some genes both in mRNA transcript and protein translation phrase. Results: TARCD could markedly improve the learning and memory abilities of all types of dementia model rats and increase the activities of PC 12 cells in vitro. It could increase the content of protein and nucleic acid, inhibit the activity of ChE, enhance the activity of SOD and reduce the content of MDA. It also increased the expression of ChaT and reduced the expressions of APP and A β . Conclusion: TARCD can improve the learning and memory abilities of dementia model rats, and its mechanisms includes improving the function of cholinergic system, facilitating the elimination of oxygen-derived free radicals, accelerating the synthesis of protein and nucleic acid in brain constitution, and reducing the deposition and toxicity of A β...
