Objective:To study the effects of Polygonum multiflorum on learning and memory ability and miRNA-101 expression in bilateral hippocampal region of Alzheimer's disease model rats,and to explore its mechanism.Methods:120 SD rats were randomly divided into 4 groups,30 rats in each group,named as sham-operation group,model group,control group and experimental group.The control group was treated with Shishan alkaloid A,and each group was given the same amount of normal saline respectively after making the model(sham operation group and model group).After 21 days of gastric perfusion,Morris water maze was used as the behavioral test method to evaluate the learning and memory ability of rats.Behavioral observation was conducted for 5 days.The experimental rats were killed at the end of the study.Bilateral hippocampal miRNA was performed with RT-qPCR.-101 semi-quantitative detection.Results:After 21 days of gavage,The learning and memory ability of the experimental group and the control group was better than that of the model group(p<0.05),and the relative expression of miRNA-101 in the bilateral hippocampal area of the experimental group and the control group was significantly higher than that of the model group(P<0.01),and the experimental group was higher than the control group(p<0.05).Conclusion:Polygonum multiflorum can significantly improve the learning and memory ability of rats with Alzheimer's disease and has a certain therapeutic effect on Alzheimer's disease.Improving symptoms by increasing the content of miRNA-101 in hippocampus of model rats may be one of its mechanisms.
