Aim: To study the effects of Compound 909 on the learning and memory of dementia model rats and its mechanisms. Methods: Y-electronic maze was used to study the behavior of dementia rats induced by D-galactose and QA. In order to investigate the mechanisms of Compound 909,the activities of AChE and SOD,and the levels of ACh, MDA and NO were determined. The changes of celluar ,and subcelluar structure , indexs of hemorrheology and the expression of APP were obse-ved by light microscope,electron microscope,blood viscometer and immunohistochemical method . MTT assay, DAPI staining,flow cytometry and RT-PCR were used to furtier investigate the mechanisms of Compound 909 on inhibiting the apoptosis of the PC12 cells injured by QA. Results: Compound 909 could remarkably improve the learning-memory abilities of dementia rats. It could inhibit the activity of AChE, increase the level ss of ACh, enhance the activity of SOD, depress the levels of MDA and NO. reduce the expression of APP, and decrease blood viscosity, plasma viscosity and indexs of erythro cyte aggregation. We also made it clear that it could inhibit the apoptosis and upregulate the expression of bcl-2 gene in the PC 12 cells injured by QA . Conclusion: Compound 909 could improve the learning-memory abilities of dementia rats, and its mechanisms might associate with improving the function of cholinergic system, facilitating the elimination of oxygen-derived free radicals, depressing the neuronal toxicity of NO , reducing the expression of APP, improving hemorrheology and inhibiting neuronal cells apoptosis.
