The purpose of this research is to discuss the probable mechanism of acupuncture treatment combined with pharmacotherapy regulating glutamatergic function and anti-free radical effect on the dementia rats due to hippocampal CA, lesions induced by quinolinic acid. So that the author can provide the clinical application of acupuncture treatment combined with pharmacotherapy with experimental evidences in treating Alzheimer's disease (AD). The thesis consists of two parts: theoretical research and experimental study.Theoretical research: the author thinks that the pathogenic mechanism of AD and memory disturbance dues to three factors as follows: deficiency of kidney-essence with exhaustion of marrow which is primal, blood stasis and phlegm stagnancy that are the secondary. The principal therapy is nourishing kidney and marrow. At the same time, the author discusses the relationship among excite amino acids, free radical and AD. This study partly provides theoretical evidences for the mechanism exploration of acupuncture treatment combined with pharmacotherapy.Experimental study: effects of three therapies of acupuncture treatment combined with pharmacotherapy, simple acupuncture treatment and treatment of Danggui Shaoyao Powder on the dementia rats due to hippocampal CA1 lesions induced by quinolinic acid were observed. The results suggest that acupuncture treatment combined with pharmacotherapy could significantly increase the level of glutamate (Glu), and theactivity of superoxide dismutase (SOD), reduce the level of lipoperoxides (LPO) and adjust the act ivi ty of NO/NOS in the hippocampus. Those effects correlate to the changes of rats' the learning and memory ability. In addition, rats in group treated by acupuncture treatment combined with pharmacotherapy show better learning and memory ability than those in other two groups. The results confirm that the probable mechanism of anti-dementia effect of acupuncture treatment combined with pharmacotherapy is associated with its in regulation of the glutamatergic function and improvement of ability of antioxidation. It prompt the resume of learning and memory ability in hippocampal CA, lesions and the improvement of dementia symptoms by their common action. Therefore acupuncture treatment combined with pharmacotherapy treating AD in clinic has certain scientific evidences and is worth being popularized widely.
