Objective: Research the mechanism of the therapy of expelling heat from Yingfen to Qifen which works in radiation-induced lung injury(RILI)by regulating the chemokine CCL3/CCR1 pathway,in order to providing new concepts and methods to clinical treatments with RILI,as well as providing experimental evidence to studying RILI in the perspective of the theory of febrile disease caused by latent Evil.Methods: The SD rat animal model of RILI was produced by using the 6MV-X ray linear accelerator(SSD=1m,dosage rate=200cgy/min);Observed the pathological changes in lung tissue of model rats treated with SangXingQingYing herbal soup;CCL3,IL-12,and IFN-γ protein levels in lung tissue were detected by Western blot;The expression of CCR1 mRNA,IL-4 mRNA were quantitatively detected by Real-time Quantitative polymerase chain reaction(RT-PCR);The expression of TGF-β1 was qualitatively detected by immunohistochemistry staining;levels of CTGF,HYP,IL-17 A,and IL-2,IL-6,IL-10 in blood were detected by enzyme-linked immunosorbent assay(ELISA).Results:(1)The pathological changes which respectively characterized by inflammation,transition from inflammation to fibrosis,and fibrosis were observed in 1-3 months,4-5 months,6-8 months after irradiation.(2)After irradiation,the expression of CCL3,CCR1 mRNA,IL-4 mRNA and TGF-β1 in lung tissue and levels of CTGF,HYP,IL-17 A,IL-6,IL-10 in blood were significantly increasing in model groups,whereas markedly decreased in treatment groups;protein levels of IL-12,IFN-γ in lung tissue and the level of IL-12 in blood were sharply decreasing in model groups,in contrast increasing obviously in treatment groups.(3)There were no differences between treatment groups witch treated with SangXingQingYing herbal soup and that lack of herb components exhibiting effects of expelling heat from Yingfen to Qifen.Conclusion:(1)The SangXingQingYing herbal soup may exhibit an anti-fibrosis effect by regulating the Th1/Th2 cytokine balance.(2)The SangXingQingYing herbal soup alleviates radiation-induced pneumonitis and subsequent fibrosis and exhibit curable effects to RILI,may regulate the Th1/Th2 cytokine balance through CCL3/CCR1 signal pathway.
