blank control group,the remaining 40 by intraperitoneal injection of one-off chain urea with streptozocin(STZ).Hydroxy benzene sulfonic acid calcium,concentration of low dosage group and high dosage group,gastric 42 days in a row,2 times a day,a 1 ml at nine o ’clock in the morning,lavage at two o ’clock in the afternoon.Flux hysteresis soup Ⅱ treatment effect is through the detection and analysis before dosing weight,fasting glucose,intraocular pressure,fundus color,OCT,ERG test indicators,etc and21 days and dosing weight,fasting glucose,intraocular pressure,and based on former weight,fasting glucose,intraocular pressure,retinal color,OCT,ERG,the eyeball.Using fluorescent microscope observed the pathological changes of retinal tissue of rats.And adopt Graphprism8 for groups of five and five groups before and after the treatment.Results:1.Results general condition shows:Blank control group rats’ s state of mind is good,soft hair colour and lustre is bright beautiful,activity,diet,mainland all normal.After the solution of 1% of the left lower quadrant abdominal injection of STZ rats model control group and droopy,hair yellow lacklustre,walk slow activity arch,eating and drinking water.Positive medicine control group,low concentration to medicine group,high dosage group compared with model control group,the symptoms have different degrees of improvement.2.Weight results show:For 21 days,compared with the blank control group,after STZ injection mold maker in the rat body weight were significantly decreased(P <0.01).Compared with model control group,each dose group no statistical difference(P > 0.05).42 days,dosing groups compared with model control group rats weight,no statistically difference(P < 0.05).3.Fasting blood glucose results show:For 21 days,compared with the blank control group,after STZ injection mold maker in rats were significantly elevated fasting glucose(P < 0.001).42 days,compared with model control group,positive medicine control group was not reduced blood sugar(P > 0.05),low concentration to blood sugar medicine group decreased significantly(P < 0.01),blood sugar high dosage group decreased significantly(P < 0.001).4.Fundus color results show:Model control group rats fundus color slants dark,DVD fuzzy boundaries,plates than reached around 0.9-1,vascular circuity,arteriovenous vessels,and visible blood vessels and mixed strewn at random,fundus visible hemangioma.Positive control group and low concentration to medicine group compared with model control group of the above all have different degrees of improvement.Compared with model control group,high dose group DVD boundary is relatively thin,cup dish than normal,the decrease in the number of retinal hemangioma,no bleeding,etc.42 days,compared with model control group,positive medicine control group plates than statistically difference(P < 0.05),plates high dosage group decreased significantly(P < 0.001).5.ERG results show:42 days,Compared with blank control group,model control group 10 layers is relatively retinal disorders,DVD visible edema around the whole layer of bump,Compared with model control group,to medicine group in the rat retina ten layers more neatly.compared with model group,high dosage group of the strength of a wave amplitude,wave amplitude b has a rising trend(P < 0.05);low concentration to medicine group and the high concentration of drug group OPs amplitude strength increased significantly(P < 0.01).6.OCT results show:42 days,compared with model control group,thickening of dosing retinal tissue has high concentration group(P < 0.05).7.Tunel method to detect apoptosis results show:The apoptotic cells compared with model control group,positive medicine control group significantly increased(P <0.001);low concentration group and high dosage group of apoptotic cells decreased significantly,with significant statistical difference(P < 0.001).8.Immunofluorescence test results of retinal VEGF in rats show:Compared with model control group,blank control group,low concentration dose group and high dosage group of VEGF expression is relatively small(P < 0.001).Conclusion:1.In STZ induced joint high fat and sugar feed model 72 h fasting blood glucose(>13.9mmol/L),copied the DM rat model.2.Number of traditional Chinese medicine compound flux hysteresis soup Ⅱ in raw radix rehmanniae,radix scrophulariae,radix ophiopogonis,fructus ligustri lucidi,ink dry lotus,rhubarb,fall blood sugar results curative effect is remarkable.3.Traditional Chinese medicine compound flux hysteresis soup Ⅱ number of non proliferative diabetic retinopathy induced by STZ rats have good therapeutic effect,Its therapeutic effect may be related to weight control,control blood sugar,improve blood flow,improve the function of DVD,retinal tissue nutrition,reduce apoptosis,reduce the new angiogenesis,inhibiting inflammation.Its main protection area may be located in the outer nuclear layer of rat retina structure. |
