Purpose: The purpose of this study was to collect and study the prescriptions of traditional Chinese medicine for the treatment of sinus bradycardia by Professor Wu Wensheng and various databases by data mining method,so as to analyze the medication rules of traditional Chinese medicine for the treatment of sinus bradycardia,and to explore the active ingredients,targets and pathways of action of high-frequency Chinese medicine for the treatment of sinus bradycardia by network pharmacology and molecular docking technology.Method:1.Patients with sinus bradycardia who were admitted to Professor Wu Wensheng’s outpatient Department of the First Affiliated Hospital of Liaoning University of Traditional Chinese Medicine from November 2021 to November 2022 and who were effectively treated with traditional Chinese medicine decoction for four weeks were collected.At the same time,according to the NRP standard,self-prepared prescriptions and Chinese-made prescriptions for the treatment of sinus bradycardia from China National Knowledge Network,Wanfang Data Knowledge Service platform and VIP Chinese periodical service platform were screened,and the outpatient and database data were standardized.WPSExcel,SPSS and other software were used to make statistics and analysis of the data,so as to summarize the medication rules of Chinese medicine in the treatment of sinus bradycardia.2.Network pharmacology and molecular interconnection studies were conducted on high-frequency Chinese medicines(cassia twigs,sundried licorice,Aconite,Astragalus membranaceus,salvia miltiorrhiza,asarum,angelica sinensis)obtained from data mining.High frequency TCM components and targets were obtained from TCMSP database,disease targets were obtained from Gene Cards database,Drug Bank database and Pharm GKB database,and drug components,drug targets and common targets of drug components were sorted out by perl software.Cytoscape3.9.0 was used to construct the network diagram of Drug Composition and Target,and String database was used to map the protein interaction network(PPI),and Cytoscape3.9.0 and R4.2.2 software were used to screen the PPI network core.R4.2.2 software was used for GO enrichment analysis and KEGG enrichment analysis.Finally,Chem3 D,SYBYL-X2.0 and other software were used to conduct molecular docking on key components and key targets for verification.Results:1.Finally,18 clinical cases were collected,and 259 literatures were in line with the literature inclusion standard,208 prescriptions were involved,193 TCM were involved,and the accumulative use frequency of TCM was 2205 times.The drugs used more than 70 times had a total of 7 flavors,which were listed in order: cassia branch,roasted licorice,Aconite,astragalus membranaceus,salvia miltiorrhiza,asarum,angelica sinensis.The properties of drugs used in the treatment of sinus bradycardia are mainly warm,with sweet,bitter and occinative taste,mainly entering the liver,spleen,lung,stomach,kidney and heart meridian.The efficacy of traditional Chinese medicine is mainly based on tonifying deficiency,relieving surface,promoting blood circulation and removing blood stasis,and warming Li.The most commonly used two drugs are asarum-ephedra,aconite-ephedra,ophiopogon-Schisandra,sundried licorice-jujube,the most common the three drugs were associated with ephedra,aconite and asarum.Two cluster prescriptions were obtained by cluster analysis,which were prescription 1: asarum,aconite,ephedra,cassia twig,schisandra chinensis,prepared licorice,ophiopogon ophiopogon and salvia miltiorrhiza.Prescription 2: Angelica sinensis,Codonopsis,Atractylodes Rhizome,Astragalus membranaceus,Ligusticum Chuanxiong.2.215 chemical components and 11098 targets of high-frequency traditional Chinese medicine were obtained by network pharmacology,among which 153 chemical components and 52 potential targets were related to the treatment of sinus bradycardia.Quercetin,7-O-methyl isomicrocoxacin,2-isopropyl-8-methylphine-3,4-dione, isocryptotanshinone and Tanshinone IIA are the key active components of high-frequency traditional Chinese medicine,and IL1 B,CASP3,VEGFA,TNF,CCL2,CXCL8 and IL6 are the key targets of high-frequency drugs in the treatment of sinus bradycardia.Involved in diabetes complications of AGE-RAGE signaling pathways,Malaria,Inflammatoryboweldisease and relevant way.The results of molecular docking showed that the key components into blood and the core target proteins could combine through hydrogen bonding and had strong bonding activity,which could form stable spatial structure.Conclusions:1.The drug properties of sinus bradycardia of Chinese medicine are mainly warm,with sweet,bitter and occinative tastes,which mainly enter the liver,spleen,lung,stomach,kidney and heart channels.The efficacy of Chinese medicine is mainly related to tonifying deficiency,relieving surface,promoting blood circulation and removing blood stasis,and warming Li.Two common prescriptions for sinus bradycardia were obtained by cluster analysis,which were ephedra aconiti asarum decoction combined with Shengmai decoction and Guipi decoction,respectively.In order to introduce,TCM treatment of sinus bradycardia focuses on nourishing blood and qi,nourishing Yin and warming Yang,Qi and removing blood stasis,nourishing blood and calming the spirit.2.The treatment of sinus bradycardia by high-frequency Chinese medicines(cassia twigley,processed liquorice,aconite,astragalus membranaceus,salvia miltiorrhiza,asarum,and Dangica sinensis)may be possible through quercetin,7-o-methyl-isopyloxacin,2-isopropyl-8-methylphenanthrene,4-dione,isocryptotanshinone,tanshinone IIA and other active ingredients.It acts on IL1 B,CASP3,VEGFA,TNF,CCL2,CXCL8,IL6 and other targets through AGE-RAGE signaling pathway in diabetes complications,malaria and other pathways.The molecular docking results showed that the key components were linked to the core target by hydrogen bonding,which could form a stable spatial structure.It provides some reference for further research. |
