| Objective: To investigate the relationship between human leukocyte antigen(HLA)-DQB1 alleles and oral submucosal fibrosis(OSF),and the effect of Modified Danxuan Koukang on the expression of differential genes,and to study the susceptibility of OSF and the intervention mechanism of Traditional Chinese Medicine from the perspective of HLA gene polymorphism,so as to provide a new basis for clinical prevention and treatment of OSF.Methods:(1)OSF patients and healthy volunteers were recruited according to inclusion and exclusion criteria and divided into OSF group and normal group.Polymerase chain react-sequence-specific primers(PCR-SSP)was used to detect the expression frequency of HLA-DQB1 alleles in the two groups and analyze the significant allelic subtypes and their relative risk.(2)Based on the identification of differential genes,real-time quantitative PCR(q PCR)was used to detect differential gene expression levels,and the gene expression levels of OSF patients and healthy volunteers expressing differential genes were compared.(3)The intervention treatment of OSF patients with differential gene expression was performed by Jiawei Dan Xuankoukang,and the expression level of the HLA-DQB1 gene in OSF patients before and after treatment was compared.Results:(1)The expression frequency of HLA-DQB1 alleles were detected in 44 OSF patients and 44 healthy volunteers.A total of 8 specific HLA-DQB1 alleles were detected.The expression frequency of the DQB1*0502 allele in the OSF group(36.4%)was significantly higher than that in the normal group(13.6%)(P < 0.05).There were no significant differences in allele expression frequencies of DQB1*0201,DQB1*0303,DQB1*0501,DQB1*0503,DQB1*0602,DQB1*0603 and DQB1*0604 in the OSF group compared with the normal group(P > 0.05).(2)HLA-DQB1*0502 allele exposure was positively correlated with oral submucosal fibrosis(OR = 3.619,P < 0.05).(3)The relative expression level of the HLA-DQB1*0502 allele in the OSF group(3.98 ± 3.50)was significantly higher than that in the normal group(0.70 ± 0.41)(P < 0.05).(4)The relative expression level of the HLA-DQB1*0502 allele in patients with OSF after TCM treatment(2.60 ± 2.08)was significantly lower than that before treatment(3.98 ± 3.50)(P <0.05).Conclusions:(1)HLA-DQB1 allele polymorphisms are different between patients with oral submucosal fibrosis and the normal population,and it is preliminarily suggested that the HLA-DQB1*0502allele may be a susceptibility gene for oral submucosal fibrosis,which has a high correlation with OSF.(2)The expression level of allele HLA-DQB1*0502 in patients with oral submucosal fibrosis was significantly higher than that in the normal population.(3)Modified Danxuankoukang can significantly reduce the expression of the HLA-DQB1*0502allele which is an important immune function indicator. |
