| Objective: To observe the inflammatory response and histopathological manifestations of anterior segment of Experimental autoimmune uveitis(EAU)rats,to evaluate the efficacy of Feng Mingmu Pill on EAU rats and the expression of key proteins of JAK/STAT pathway in EAU rat retinal tissue,and to explore the possible mechanism of action of Feng Mingmu Pill in EAU rats.Methods: Seventy SPF grade male Lewis rats(140 eyes)were randomly selected as the blank group according to the random number table method,and the remaining were used as the modeling group for active immunization.Lewis rats were immunized subcutaneously with Fre und’s complete adjuvant(CFA)chyle solution containing inter photoreceptor retinoid binding protein(IRBP)on the inner side of the hind legs for 4 consecutive days,Establish an EAU rat model.On the first day after immunization,the inflammatory manifestations of the anterior segment of the rats were observed,and the EAU rats with successful mo deling were divided into model control group,low-dose group and normal dose group of Wind Removal Pill(hereinafter referred to as control group,low-dose group,and normal dose group)according to the random number table method,with 16 rats in each grou p.Each group of rats is dosed as planned.On the 7th day after molding and the 14 th day after gavage,the correspon ding rat retinal tissue was stained for HE staining t o make pathological sections for histopathological examination.Western Blot detection was used to detect the relative expression of JAK,STAT1 and STAT3 proteins in rat retinal tissues.Results: 1.Histopathological results showed that on the 7th day afte r immunization,the pathological score of tissue sections in the model group was higher than that in the blank group,and the EAU rat model was successfully established.On the 14 th day after administrat ion,the degree of retinal tissue structure destructi on in the intervention group was significantly lighter than that in the model group,indicating that the drug intervention had an effect.2.The results of Wb detection showed that compared with the blank control group,the model control group had high exp ression of JAK,STAT1 and STAT3,and the results were statistically significant(P<0.05);Compared with the model control group,the expression of JAK,STAT1 and STAT3 in the intervention group wa s statistically significant(P<0.05).Conclusion:1.Wind dispelling Mingmu Pill can downregulate the expression of JAK,STAT1 and STAT3 in EAU rat retinal tissue,indicating that JAK,STAT1 and STAT3 may be the targets of Wind Removal Pill acting on EAU rats,wh ich can improve the pathological process of EAU by in hibiting the above targets.2.Wind dispelling Mingmu pill may exert its anti-inflammatory effect by inhibiting the expression of JAK,STAT1 and STAT3 proteins in the JAK/STAT signaling pathway that causes uveitis. |
