Objective: To establish the model of retinal light damage in Sprague-Dawley rats and observe the changes of pathological morphology,thickness of outer nuclear layer and apoptosis index,to investigate the protective effect of Ziyinmingmu pill on light induced retinal damage in rats.Methods: 60 Sprague-Dawley(SD)rats were randomly divided into 6 groups: control group,model control group,Qijudihuang pill(positive control group),Ziyinmingmu pill low dose group(low dose group),Ziyinmingmu pill in dose group(middle dose group),Ziyinmingmu pill high dose group(high dose group n = 10).All SD rats were adapted for 10 days in 12h(20~50Lux)and 12 h dark(0~12Lux)cycle light and white fluorescent lamps.Except control group,other 5 groups were established in rats model of retinal light injury,light injury 3 days after intragastric gavage for 15 days,the rats were killed and the eyes were enucleated,preparation of paraffin section and HE staining,microscope observation of retinal pathological changes,measurement of retinal thickness of the outer nuclear layer after,apoptosis by TUNEL markers as sections were observed under the light microscope,measuring cell apoptosis index.Results:1.HE staining: blank control group retina hierarchy,structure clear and complete,uniform dyeing,the outer nuclear layer(outer nuclear layer,ONL)arranged in regular cell shape,nuclear staining deep,outer nuclear layer thickness(54.59 ± 7.42)?m model control group;each retinal layer structure fuzzy boundaries.Not clear,ONL cells reduced,arranged in disorder,thinner,part of the nuclear staining fades,the thickness of the outer nuclear layer is(16.64 ± 5.76)?m,compared with the blank control group,P<0.01(P=0.000);low dose group retinal structure was disordered,the outer nuclear layer thickness is thin,outer core the layer thickness is(17.97 ± 6.52)?m,compared with the model group,P>0.05(P=0.446);middle dose,high dose group,positive control group,the retinal structure aligned in order,level is clear,the thickness of the outer nuclear layer increases,and the blank control group Comparison of P values were greater than 0.05,compared with the model group,P<0.01,there was no significant difference between the three groups(middle dose group and high dose group,P=0.965,middle dose group and positive control group,P=0.739,high dose group and positive control group,P=0.758).2.TUNEL staining: In the blank control group,there were no nuclei in the outer nuclear layer;the model control group in the outer nuclear layer of retina positive nuclei were labeled positive nuclei labeled brownish yellow nuclei is not uniform,the cell membrane integrity,chromatin condensation into "crescent nucleus" or "ring shaped nuclear,nuclear chromatin condensed in the formation of the" eyeball nucleation "in the middle;the marked reduction of nuclei in low dose group of retinal outer nuclear layer;labeled nuclei in the outer nuclear layer,high dose group and positive control group was significantly reduced.The control group AI was 1.5439% ± 1.5154%,AI in model group was much higher than that of control group(P < 0.01),61.0855 ± 4.9529%,the low dose group AI = 58.7048 ± 7.1547%,and the model control group(P=0.527);The AI values of the middle and high dose group and positive control group were significantly lower than that of the model group(P < 0.01).There was no significant difference between the three groups(P > 0.05).Conclusions: Ziyinmingmu pill can increase the thickness of the outer nuclear layer of rat retinal light damage,inhibit the apoptosis of light injury in rats,the rat retinal light injury has protective effect.
