Object:To observe the effect of QufengHuoxue pill on experimental autoimmune uveitis(EAU)rat serum IL-4?IL-15 and IFN-?.Method: Eighty(160 eyes)male Lewis rats were randomly selected according to the random number table method as A blank group(A),and the remaining 64 rats were respectively injected with Complete freund's adjuvant(CFA)CD of inter-photoreceptor retinoid binding protein(IRBP)1177-1191.Lewis rats were actively immunized,and A EAU rat model was established.After modeling,the inflammation in anterior ganglia of rats was observed under slit lamp microscope,and the inflammation was scored according to Caspi scoring criteria.On the 7th day after immunization,Lewis rats' eyeballs were taken for pathological sections,and histopathological examination was conducted after HE staining.After the model was completed,it was randomly divided into model group(B),QufengHuoxue pill routine dose group(C),QufengHuoxue pill double dose group(D),and XiongdanKaiming tablet group(E)according to the random number table method.The relative expressions of IL-4?IL-15 and IFN-? in serum of rats were detected by ELISA method after 14 days of continuous gavage.Results:1.Verify the EAU rat model: the inflammatory reaction of the anterior segment was observed under the slit lamp microscope on the 4th day after the immunization of the rats in the model group,which showed that the iris blood vessels of the rats were slightly dilated,the blood vessels of the limbus of the cornea were congested,and the pupils were narrowed;Eye inflammation score(0.30±0.21).On the 5th day,there were various degrees of anterior chamber turbidity,posterior corneal wall apoptosis(KP),aqueous flash,pupil constriction but still visible pupil,eye inflammation score(0.47±0.09).On day 6-7,significant uveitis reaction was observed,which was characterized by severe anterior chamber turbidity(empyema),aqueous turbidity,greyish flocculent at the pupil margin,pupillary membrane occlusion,and eye inflammation score(2.06±0.68,2.81±0.75).On the 7th day after immunization,pathological sections of the rat's eyeball showed different degrees of inflammatory cell infiltration in the cornea,iris root,ciliary body,retina and other parts of the rat.The above results indicated that the EAU rat model was successfully established.2.ELISA results showed that IL-4?IL-15 and IFN-? were significantly increased at each time point in the model group compared with the blank group,and the difference was statistically significant(P<0.01).The expression of IL-4: compared with the model group,the expression of QufengHuoxue pill in the conventional dose group,QufengHuoxue pill in the double dose group and XiongdanKaiming tablet group were significantly increased,with statistically significant differences(P<0.01).The expression of IL-15: compared with the model group,the expression of QufengHuoxue pill in the routine dose group was decreased,and the difference was statistically significant(P<0.05).The expression of QufengHuoxue pill was significantly decreased in the double-dose group and XiongdanKaiming tablet group,and the difference was statistically significant(P<0.01).The expression of IFN-?: compared with the model group,the expression of QufengHuoxue pill in the conventional dose group,QufengHuoxue pill in the double dose group and XiongdanKaiming tablet group was significantly decreased,with statistically significant difference(P<0.01).Conclusion:1.Lewis rats were actively immunized with IRBP antigen-adjuvant emulsifier,and the rat model was successfully induced and established.2.QufengHuoxue pill can inhibit the expression of IL-15 and IFN-? in EAU rats,and enhance the expression of IL-4 in EAU rats.It is confirmed that IL-4?IL-15 and IFN-? are the targets for intervention of of QufengHuoxue pill intervention against EAU.Prove that functions of QufengHuoxue pill can inhibit the expression of IL-15 and IFN-?,enhance the expression of IL-4,promote to achieve the purpose of improving EAU pathological process.3.QufengHuoxue pill may exert its anti-inflammatory effect by inhibiting the expression of pro-inflammatory cytokines IL-15 and IFN-? and enhancing the expression of anti-inflammatory cytokines IL-4.
