Prunus salicina ’Fengtangli’,a genus of plum in the Rosaceae family,was currently mainly planted in several areas such as Zhenning County and Ziyun County.It has been named as a Chinese National Geographical Indication protected product,and has been selected for the National List of Famous,Special and New Agricultural Products,etc.The ’Fengtangli’ has long been an important agricultural industry and a key project in the fight against poverty in Zhenning County,Anshun City,But with the increase of ’Fengtangli’ cultivation area,the ’Fengtangli’ seedling market is in disarray and the problem of the lack of a parent source of fruit seedlings is becoming increasingly prominent.Therefore,the establishment of a reliable technical system for the identification of ’Fengtangli’ is particularly important for the maintenance of order in the ’Fengtangli’ market and the healthy development of the ’Fengtangli’ industry.In this paper,72 plum germplasm collections(including 52 ’Fengtangli’ as the experimental group and 20 other plum varieties as the control group)were used as experimental materials to establish the identification system of ’Fengtangli’ and to analyse the affinity between ’Fengtangli’ and 20 plum varieties using morphological marker techniques and two molecular markers(ISSR and SSR).The aim was to provide assistance for the healthy development of the ’Fengtangli’ industry.The main findings of the study are as follows:1.A total of 72 samples were tested for 14 fruit physiological indicators.Combining the main intrinsic fruit indicators,it was found that the soluble solids content,sugar content and Vc content of ’Fengtangli’ were significantly higher than those of other plum varieties.A principal component analysis of the 14 physiological indicators revealed that ’Fengtangli’ had the first highest overall score among the 21 plum varieties,finding that the fruit quality of ’Fengtangli’ was better than most of the plum varieties offered for trial.When the varieties were identified based on fruit traits,errors were found in the clustering results,but most of the plums could still be identified.Molecular marker studies should also be carried out to identify the plums more accurately.2.The 10 core primers were selected from 100 ISSR primers published by Columbia University,and the average PIC of the 10 core primers was 0.8115.80 polymorphic loci were amplified in 72 samples,with an average polymorphic locus ratio of 97.14%.The DNA fingerprints of the 72 samples were constructed,and the 10 core primers were able to distinguish the 52 plums from the 20 plums in the control group.It was found that ’Fengtangli’ was more closely related to Qingcuili plum and’Fengtangli’ was more distantly related to Dahongpao.But,no specific loci were found in the 10 core primers amplification sites,so more efficient molecular markers need to be introduced to identify ’Fengtangli’.3.18 pairs of SSR core primers with high polymorphism were screened,and the mean PIC value of the 18 pairs was 0.8161.365 polymorphic loci were detected in 72 test samples,and the range of polymorphic loci was from 13 to 33.A technical system based on SSR markers was established for the identification of ’Fengtangli’,and SSR fingerprint profiles were constructed for the 72 test materials.The clustering results were similar to those of ISSR.And the core primers screened had universal characteristic loci in ’Fengtangli’.Therefore,the identification system and SSR fingerprinting can provide strongly supported for the identification of ’Fengtangli’.4.The genetic distances of the three markers(ISSR,SSR and fruit traits)were analysed by Manel test.The results showed that all three markers were highly significantly correlated with each other,indicating that the three markers were related to each other to some extent and showed consistency in terms of genetic similarity.And the efficiency of identification of ’Fengtangli’ was SSR>ISSR>morphology.
