Identification of seed authenticity and purity is critical for assurance seed quality,the use of DNA markers to detect the quality of the seed,not only fast and efficient,and more accurate.In this study,15 large-scale cultivation hybrid cotton varieties purchased by seed market are taken as materials,multiple DNA molecular marker is used for their variety identification and purity analysis,try to find the technology system that do not rely on two parents market,fast,high-throughput and economy to identify F1 The main results are described as follows:(1)Five specific amplification primers are screened from ten ISSR primers,and through the test of the annealing temperature and cycle times,the applicable amplification procedures of the five ISSR primers are established.Will these 5 ISSR primers used in 15 samples of cotton PCR amplification,by polyacrylamide electrophoresis to detect the specific bands,11 samples can be completely separate from the 15 samples.By using DPS software clustering analysis,the results show that the genetic relationships of the 15 cotton cultivars are close.(2)Comparing the practicability of TRAP,SRAP and ISSR molecular markers in the Cultivar identification of the selected Cotton samples,the ISSR molecular marker owns the best effect in this experiment,and the specific bands most abundant,stable,and the most significant.(3)112 grains of one sample(Si hybrid cotton 8 F1)are selected to extract genome DNA,and the genome DNA are taken as the template for ISSR-PCR amplification,then by silver staining the purity of the breeds are detected.In this study,the conclusion: The ratio that consistent with the number of samples band type is 79.5%.So speculate purity of the sample was only 79.5%.But the authenticity and purity of the breed need further planting,by morphological identification results indicators and other indicators to confirm each other,but also help to verify the efficiency of molecular markers in order to obtain a final conclusion.Or after obtaining the two parental species,we can identify F1 and them at the same time.
