Objective:To investigate the expression and correlation of nucleobindin-1(NUCB-1)and COX-2 in colorectal adenocarcinoma.Methods:1.The STRING database was used to screen out the molecules related to COX-2,and UALCAN,Kaplan-Meier Plotter,and the HPA database predicted COX-2 and NUCB-1 expression and its impact on colorectal adenocarcinoma prognosis.2.The eligible cases of colorectal cancer from June 2019 to June 2020 were collected from the Department of Gastrointestinal Surgery of the First Affiliated Hospital of Gannan Medical University.The expression of NUCB-1 and COX-2 in cancer tissues and adjacent tissues was detected by immunohistochemistry.The chi-square test was used to verify whether the expression of these two molecules was statistically significant and whether it was related to the degree of differentiation,clinical stage,lymphatic metastases,and other clinical parameters,and then the Spearman test was used to verify the correlation between the two molecules.Results:1.Bioinformatic analysis:the expression of COX-2 in colorectal adenocarcinoma was significantly increased,especially in stages II and III,and COX-2 was associated with poor prognosis.With the help of the STRING database,molecules closely related to COX-2 were screened out,and NUCB-1 was finally determined to be included in the study.Expression of NUCB-1 was reduced in colorectal adenocarcinoma.Paradoxically,high expression NUCB-1 was associated with shorter RFS and OS time.2.Immunohistochemistry:(1)COX-2:in cancer tissues,COX-2 was expressed in 39 cases and negative in17 cases,with a positive rate of 69.64%.There were 16 cases of positive expression and 40 cases of negative expression in adjacent tissues,with a positive expression rate of 28.57%.There was a significant difference in the expression of COX-2 between cancer tissues and adjacent tissues(X~2=19.85,P<0.005).There were significant differences between the expression of COX-2 and clinical stage(X~2=9.80,P<0.050),degree of differentiation(X~2=8.18,P<0.050),and lymphatic metastases(X~2=4.72,P<0.050),but not with age(X~2=0.28,P>0.500),gender(X~2=2.48,P>0.100),distant metastasis(X~2=1.87,P>0.100)and degree of invasion(X~2=1.35,P>0.100).(2)NUCB-1:it was found that there were 36 cases of positive expression and 20cases of negative expression in cancer tissue,with the positive expression rate being64.29%.There were 11 cases of positive expression and 45 cases of negative expression in adjacent normal tissues,with a positive expression rate of 19.64%.There was a significant difference in the expression of NUCB-1 between cancer tissues and adjacent tissues(X~2=24.79,P<0.005).There were significant differences between the expression of NUCB-1 and clinical stage(X~2=9.18,P<0.050),degree of differentiation(X~2=6.89,P<0.050),and lymphatic metastases(X~2=4.07,P<0.050),but not with age(X~2=0.04,P>0.500),gender(X~2=2.07,P>0.100),distant metastasis(X~2=2.40,P>0.100)and degree of invasion(X~2=2.11,P>0.100).(3)Correlation analysis of NUCB-1 and COX-2:Spearman test analysis showed that the correlation coefficient between COX-2 and NUCB-1 was r_s=0.6312 and P<0.001.Conclusion:1.Compared with adjacent normal tissues,the expression of COX-2 in colorectal adenocarcinoma is significantly up-regulated and correlated with clinical stage,degree of differentiation,and lymphatic metastases.2.Compared with adjacent normal tissues,the expression of NUCB-1 in colorectal adenocarcinoma is significantly up-regulated and correlated with clinical stage,degree of differentiation,and lymphatic metastases.3.There is a significant strong positive correlation between COX-2 and NUCB-1in colorectal adenocarcinoma.
