Objectives To investigate the relationship between LCE1 C gene polymorphisms and psoriasis vulgaris.Methods In this study,a total of 101 psoriasis patients and 79 healthy controls were subjected to exome sequencing.LCE1 C gene and the regions of upstream and downstream 2kb were sequenced.Single nucleotide polymorphism(SNP)based association analysis and haplotype-based association analysis were performed in SNPs with a minimum allele frequency greater than 1%.Bioinformatics methods were used to predict the impact of risk loci on gene function.Results By sequencing the exon region of the LCE1 C gene and its upstream a nd downstream 2 kb,9 SNPs were discovered.SNP-based association analyses showed that 4 SNPs(1 in 5’UTR,1 in exon 2,and 2 in 3’UTR)were suscep tible to psoriasis vulgaris(OR=0.284~3.742,95%CI:0.086~13.630,P<0.05).Bioi nformatic prediction indicates that rs2006940,located in exon 2,can convert c ysteine to tyrosine.Haplotype-based association analyses showed that 1 haplot ype rs150288174/rs377258958/rs200887064--G was the protective haplotype of disease(OR=0.284,95%CI:0.085~0.944,P=0.040),and the frequencies of the haplotype was 6.33% in controls and 1.98% in patients.Conclusions LCE1 C gene is associated with the susceptibility of psoriasis in Hainan Han population.
