Study On The Inhibition Of Malignant Phenotype Of Head And Neck Squamous Cell Carcinoma By Wall Removing Ingredients Of Ganoderma Lucidum Spore Powder And Its Molecular Mechanism

Objective To investigate the effect of Ganoderma lucidum spore powder on the malignant phenotype of SCC15 and SCC25 cells and its molecular mechanism.Method(1)Calculates the IC50 of each time period.(2)The clone formation experiment was conducted to observe and compare whether Ganoderma lucidum spore powder affected the clone formation ability of SCC15 and SCC25 cells.(3)Scratch test was conducted to investigate whether Ganoderma lucidum spore powder affected the mobility of SCC15 and SCC25 cells.(4)The effects of Ganoderma lucidum spore powder on the metastasis of SCC15 and SCC25 cells were investigated in Transwell chamber.(5)The effect of Ganoderma lucidum spore powder on the apoptosis of SCC15 and SCC25 cells was detected by flow cytometry.(6)The effect of Ganoderma lucidum spore powder on cell cycle distribution was detected by flow cytometry.(7)WB was used to determine whether Ganoderma lucidum spore powder affected the expression of apoptosis-related proteins.(8)Changes of EMT-related markers were detected by WB.(9)WB was used to detect the expression of cell cycle-related proteins.Results(1)The IC50 values of Ganoderma lucidum spore powder in SCC15 cell line for 24 h,48h and 72 h were 1460ug/ml,501ug/ml and 189.7ug/ml,respectively.(2)The IC50 values of Ganoderma lucidum spore powder in SCC25 cell line for 24 h,48h and 72h were 1460ug/ml,1060ug/ml and 548.8ug/ml,respectively.(3)Ganoderma lucidum spore powder inhibit tumor cell metastasis.(4)Ganoderma lucidum spore powder induce cell apoptosis and block cell cycle.(5)Ganoderma lucidum spore powder inhibit EMT.(6)Ganoderma lucidum spore powder up-regulated p21 and p27,down-regulated CDK1.Conclusion(1)Ganoderma lucidum spore powder can inhibit the proliferation and metastasis of SCC15 and SCC25 cells,and promote apoptosis.(2)Ganoderma lucidum spore powder inhibits EMT.(3)Ganoderma lucidum spore powder may stagnate in the G2/M phase through up-regulation of p21,p27 and down-regulation of CDK1.
