| Ganoderma has a variety of nutrients and many kinds of biological activities & pharmacological effects, therefore a series of functional food or medicine are developed with the Ganoderma lucidum spore powder as raw material is growing, but so far there are no official quality standards for Ganoderma lucidum spore powder, and the researches to Ganoderma's quality are more concentrated in quality control of Ganoderma lucidum polysaccharides and Ganoderma acid (triterpenoids), While adenosine, as one of its main active substance, its research is less. This research builds up the measuring method of adenosine content,Ganoderma lucidum polysaccharides in Ganoderma spore powder and a high-performance liquid chromatography fingerprints with the nucleoside substances as reference materials.Chromatography can reflect the Ganoder's overall features with the nucleoside as an indicator, providing in-depth identification basis to the evaluation of the quality .1. The study on determination of active ingredients in Ganoderma lucidum spore powderObjective: The establishment of adenosine content and Ganoderma lucidum polysaccharide on Ganoderma lucidum spore powder in sample .And using them as indicators of quality control of Ganoderma lucidum spore powder.Methods: With 10% methanol solvent, ultrasonic extraction 50min, 0.01mol / L sodium acetate solution: methanol = 85:15 (V / V) as mobile phase, UltimateTM AQ-C18 (4.6×250mm, 5μm) column, 260nm detection wavelength, HPLC-PDA determination of adenosine; using UV spectrophotometry, sulfuric acid - anthrone color method in 625nm wavelength absorbance values are measured to determine the content of Ganoderma lucidum polysaccharides. Results:Adenosine content of Ganoderma lucidum spore powder reaches 258.42μg/g, the average recoveries were between 98.04% to 99.64%; Ganoderma lucidum polysaccharide content reaches 1.75%, the average recoveries are between 82.83% to 90.00%, which can meet the analytical requirements of determination of adenosine and Ganoderma lucidum polysaccharide.Couclusion:The proposed method has highly repeatability and selectivity,it is simple and fast.2.The study on fingerprint by high performance liquid chromatographyObjective:The establishment of high performance liquid chromatographic fingerprint of Ganoderma lucidum spore powder which take adenosine, uridine as reference materials.Methods: With 10% methanol solvent,ultrasonic extraction 30min,0.01mol / L sodium acetate solution–methanol as mobile phase gradient elution, UltimateTM (4.6×250mm, 5μm) column, 260nm wavelength detection, HPLC-PDA method.Results:The similarity of S1~S9 are between 0.925 to 0.947;The similarity of S10 is 0.827.Couclusion: The established HPLC fingerprint has highly repeatability and selectivity,it is simple and fast,which can provide a deeper evaluation of the quality of Ganoderma lucidum spore powder. |
