Objective: To study the Protection by Ganoderma lucidum spore powder against liver injury in subchronic cadmium exposed rats and for feasible protecting approach.Methods: A total of 84 healthy male Sprague-Dawley(SD)rats,weighing 90±10g,were randomly divided into 4 groups(n=21),including normal control group(C),cadmium alone group(Cd)and two doses of Spore powder groups(GCd L and GCd H groups).After one week adaptation treated with general meals and diet balance,Cd Cl2(1.0mg/kg)were given to Cd group,GCd L group and GCd H group via intraperitoneal injection.Ganoderma lucidum spore(0.5 g / kg,1.0 g / kg + Cd Cl2)were given to GCd L group and GCd H group via gavage,while the same dose of distilled water were given to C group and Cd group,respectively.All rats were weighed every 7 days with cognitive performance observing.Rats were sacrificed in three batches,7 rats in each batch,on day 30,60 and 90 respectively.Blood sample were taken from the carotid arteries.The liver was harvested rapidly and irrigated by normal saline.Filter paper was adopted to wipe the rest of water off.All samples were weighted,recorded and stored at-80℃ before detection.The serum alanine aminotransferase(ALT)and serum the content of IL-6 in liver were detected by enzyme-linked immunosorbent assay(ELISA).Real time RT-PCR was used to detect the content of MDA.The content of MDA and the expression of p65 m RNA,Mt-1 m RNA and Mt-2m RNA was detected by colorimetry and optical microscopy respectively.Results: The body weight of the rats in Cd group was significantly lower than C group(P<0.05).After 90 days of treatment,the body weight of the GCd H group was higher than that of Cd group(P<0.05).Compared with C group,the organ coefficient of the rats was significantly higher after cadmium staining(P<0.05).The liver organ coefficient of the rats in GCd L and GCd H was significantly higher than that of Cd group(P<0.05).Compared with C group,the levels of ALT and AST in the Cd group were significantly higher(P<0.05),while the levels of ALT and AST in Ganoderma lucidum spore powder decreased in various degree,and the difference between GCd L and GCd H was significant(P<0.05).Histopathological observation showed: intercellular substance congestion,cluttered cell line and unclear cell boundaries were ubiquitous in Cd group.While,such damages were reduced in GCd L group and GCd H group,in which GCd H group was superior to GCd L group.The content of MDA in Cd group,GCd L group and GCd H group was significantly higher than that of C group(P<0.05).The content of MDA deceased from 60 to 90 days in GCd L group and GCd H group(P<0.05).In 30,60 and 90 days,the levels of IL-6(IL-6)of Cd group were significantly high than C group(P<0.05).Compared with the Cd group,the expression of Mt-1 m RNA and Mt-2 m RNA in the GCd L group and the GCd H group rose with time(P<0.05)but the expression of p65 m RNA significantly decreased(P<0.05).Conclusion: Ganoderma lucidum spore powder has protective effect on subchronic cadmium exposed rats.And some of the underlying mechanisms may be related with antioxidant stress,reducing expression of IL-6 and inducing MT production.
