| Objective:To explore the expression and significance of FAM60A,β-catenin and c-MYC in colorectal tubular adenomas and colorectal adenocarcinomas,and analyze the correlation among them,and further study the relationship between their combined expressions and clinicopathological features,and the influence on the prognosis of patients.Methods:The m RNA expression of SIN3A/HDAC complex subunit in different types of cancer was analyzed by Oncomine database(https://www.oncomine.org).The expression level of FAM60A and c-MYC in colorectal cancer and the correlation between FAM60A and c-MYC were queried by GEPIA database(http://gepia.cancer-pku.cn/detail.php).We collected low-grade intraepithelial neoplasia tubular adenoma(n=50),high-grade intraepithelial neoplasia tubular adenoma(n=50),early colorectal adenocarcinoma(n=64)and advanced colorectal adenocarcinoma(n=131),and 75 cases of normal colorectal mucosa tissues from Gansu Provincial Cancer Hospital and Lanzhou University First Hospital from January,2013 to December,2021,underwent radical surgical resection of colorectal tumors.Made tissue microarray and detected the expression of FAM60A,β-catenin and c-MYC in each group by immunohistochemical method of SP.IBM SPSS 26.0software was used for data analysis.Result:1.Through Oncomine database analysis,it was found that the expression of FAM60A was significantly increased in colorectal cancer.Through GEPIA database,it was found that the expression of FAM60A and c-MYC in colorectal cancer was significantly higher than that in normal tissues(P<0.05),and showed a positive correlation between their expressions.2.IHC results showed that the positive expression rate of FAM60A increased gradually from normal colorectal mucosal tissue,low grade intraepithelial neoplasia tubular adenoma,high grade intraepithelial neoplasia tubular adenoma,early colorectal adenocarcinoma,advanced colorectal adenocarcinoma,and the difference was statistically significant(P<0.05);The expression of FAM60A was not only significantly correlated with infiltration depth(P=0.041),but also positively correlated with Ki67 proliferation index(r_s=0.189,P=0.008).Univariate survival analysis showed that FAM60A expression was significantly correlated with progression free survival(P=0.010)and overall survival(P=0.012);multivariate survival analysis showed that FAM60A expression and infiltration depth were independent factors affecting the prognosis of colorectal adenocarcinoma patients.3.IHC results showed thatβ-catenin was expressed in cell membrane but not in cell nucleus in normal colorectal mucosal tissues and low-grade intraepithelial neoplasia tubular adenoma,the nuclear expression ofβ-catenin was gradually increased in high-grade intraepithelial neoplasia tubular adenoma,early colorectal adenoma and advanced colorectal adenoma,and the difference was statistically significant(P<0.05).The nuclear expression ofβ-catenin was significantly correlated with tumor size(P=0.016),distant metastasis(P=0.008),and Dukes stage(P=0.015).Multivariate survival analysis showed that infiltration depth was an independent factor affecting the prognosis of colorectal adenocarcinoma patients.4.IHC results showed that the positive expression rate of c-MYC increased gradually from normal colorectal mucosal tissue,low grade intraepithelial neoplasia tubular adenoma,high grade intraepithelial neoplasia tubular adenoma,early colorectal adenocarcinoma,advanced colorectal adenocarcinoma,and the difference was statistically significant(P<0.05).c-MYC expression was positively correlated with Ki67 proliferation index(r_s=0.210,P=0.003).Univariate survival analysis showed that c-MYC expression was significantly correlated with progression free survival(P=0.031),but there was no significant difference between c-MYC expression and overall survival(P>0.05);multivariate survival analysis showed that infiltration depth was an independent factor affecting prognosis of colorectal adenocarcinoma patients.5.FAM60A andβ-catenin expression were positively correlated(r_s=0.175,P=0.015),and FAM60A and c-MYC expression were positively correlated(r_s=0.415,P<0.001),andβ-catenin and c-MYC expression were positively correlated(r_s=0.313,P<0.001).The co-expression of FAM60A withβ-catenin,β-catenin with c-MYC showed that the distant metastasis rate and the proportion of stage C and stage D in Dukes stage were significantly higher than none,and the difference was statistically significant(P<0.05).In addition,the proportion of Ki67 proliferation index≥50%in co-expression group was significantly higher than that none,and the difference was statistically significant(P<0.05).6.To analyze the relationship between the combined expression of FAM60A,β-catenin and c-MYC and the prognosis,the results showed that those who were negative for all three had the best prognosis,while those who were all positive had the worst prognosis,and the difference was statistically significant(P<0.05).Conclusion:1.The expression of FAM60A,β-catenin and c-MYC in colorectal adenocarcinoma is significantly increased,which may play a role in promoting cancer.2.There is a positive correlation among the expression of FAM60A,β-catenin and c-MYC,which may play a role in the proliferation,invasiveness and distant metastasis of colorectal adenocarcinoma.3.The combined detection of FAM60A,β-catenin and c-MYC is more helpful to evaluate the prognosis of colorectal adenocarcinoma. |
