| Objective:To research the difference of circular RNAhsa＿circ＿0001785 expression between gastric cancer and its adjacent tissues,and to further explore the relationship between the expression level of the factor and different Lauren types of gastric cancer and other clinicopathological features such as sex,age,tumor diameter,lymph node metastasis,vascular tumor thrombus and nerve invasion.Methods:1.54 cases of gastric cancer pathologically diagnosed as total gastrectomy and distal gastrectomy were collected in our hospital from July2020 to July 2022,and the normal gastric tissues adjacent to the tumor above5 cm were taken as experimental control.The pathological data of patients were obtained by consulting the pathological diagnosis system,and combined with the Lauren classification criteria of gastric cancer on page127 of Clinical Pathology,the collected cases were divided into three groups:intestinal type(18 cases),diffuse type(18 cases)and mixed type(18 cases).2.The base arrangement sequence of circular RNAhsa＿circ＿0001785gene was called up in the database and handed over to Guangzhou Branch of Suzhou Jinweizhi(Anshengda)Biotechnology Limited Company for primer and internal reference sequence synthesis.Finally,according to the detailed steps in the product manual of each reagent,the experiments of RNA extraction,reverse transcription synthesis of c DNA first strand and Quantitative Real-time PCR were completed in turn,and the integrity of RNA extracted products was determined by agarose gel electrophoresis.3.QRT-PCR technique was used to detect the expression of RNAhsa＿circ＿0001785 in cancer tissues and adjacent normal gastric tissues,and to interpret the differentiation of expression between the two.To further analyze the expression difference of the factor in three different Lauren types of gastric cancer,and to study the relationship between the expression and the clinicopathological features such as age,sex,tumor diameter,lymph node metastasis,vascular tumor thrombus and nerve invasion,so as to explore the possibility of the factor as a diagnostic marker of gastric cancer.Results:1.Through the detection of RNAhsa＿circ＿0001785 expression level in54 cases of gastric cancer and its adjacent normal tissues,the research findings of 36 cases can be used for statistical calculation.The calculation conclution indicated that the expression level of RNAhsa＿circ＿0001785 was low in 32 cases of gastric cancer,26 of these low expression samples showed that the down-regulated expression was more than 2 times,the results of the other 6 cases were not more than twice;At the same time,the expression level of RNAhsa＿circ＿0001785 in 4 cases of gastric cancer was higher than that in normal gastric tissues,2 cases were up-regulated by more than 2 times,and the results of the others were not more than twice.Statistical analysis showed that the expression of RNAhsa＿circ＿0001785 was down-regulated in gastric cancer,that is,low expression(P < 0.05).2.There was no significant correlation between the expression of RNA hsa＿circ＿0001785 and different Lauren types of gastric cancer,so the results were not statistically significant(P > 0.05).3.There was no statistical significance between the age of patients,the clinicopathological features of vascular tumor thrombus and nerve invasion and the expression of RNAhsa＿circ＿0001785(P > 0.05).But there was significant difference between the sex of patients,the diameter of tumor,lymph node metastasis and the expression of RNAhsa＿circ＿0001785.(P <0.05).4.According to the analysis of gel electrophoresis experiment,RNA extracted products has good integrity and can be used in follow-up experiments.Conclusions:The expression of RNAhsa＿circ＿0001785 was down-regulated in gastric cancer,that is,the expression level of RNAhsa＿circ＿0001785 in gastric cancer was lower than that in its adjacent tissues.There was no significant correlation between the expression of RNAhsa＿circ＿0001785 and different Lauren types of gastric cancer;and there was no correlation between the expression level of RNAhsa＿circ＿0001785 and the clinicopathological features of patients with gastric cancer,such as age,vascular tumor thrombus and nerve invasion;the expression level of RNAhsa＿circ＿0001785 was correlated with sex,tumor diameter and lymph node metastasis. |
