Objective:A mounting number of circular RNAs(circRNAs)have been confirmed to have an important role in multiple cancers due to the advance of sequencing and experimental technology.We further investigated the critical function of circRNA hsacirc0005284 in gastric cancer(GC),confirmed its probable molecular mechanism,and explored its potential as a molecular biomarker in this paper.Methods:To begin,gene chip technology was performed to analyze circRNA expression in GC tissue in order to identify the index with differential expression.Second,using a qRT-PCR assay,the differential expression level of hsacirc0005284 in GC tissues and adjacent non-tumor tissues was validated.The association between the expression of hsacirc0005284 and clinicopathological features was then investigated using qRT-PCR detection results,as well as its potential as a molecular marker.Then,using a lentiviral system and puromycin screening,the hsacirc0005284 overexpression cell line was established for subsequent functional studies.CCK-8 and colony formation experiments were used to investigate the effect of hsacirc0005284 on GC cell proliferation;scratch tests were used to investigate the effect of hsacirc0005284 on GC cell migration;and the Transwell test was used to investigate the effect of hsacirc0005284 on GC cell invasion.Finally,Western Blot was used to confirm the putative molecular control mechanism of hsacirc0005284.Results:The expression level of hsacirc0005284 was higher in GC tissues than adjacent non-tumor tissues,according to gene chip and RT-PCR data.Furthermore,we discovered that hsacirc0005284 expression was linked to sex,but not to patient age,tumor size,TNM stage,vascular invasion,neural invasion,or American Joint Committee on Cancer(AJCC)stage.Overexpressed hsacirc0005284 promoted the proliferation,migration,and invasion of GC cells in CCK-8,colony formation,scratch healing assay,and Transwell assays.Overexpressed hsacirc0005284 boosted the up-regulation of EMT-related proteins Ncadherin and Vimentin,as well as the down-regulation of E-cadherin,according to Western Blot tests.Conclusion:In the tissues of GC patients,hsacirc0005284 was overexpressed.Functionally,the overexpressed hsacirc0005284 promoted cancer by stimulating the proliferation,migration,and invasion of GC cells.Mechanistically,overexpressed hsacirc0005284 upregulated the expression level of EMT-related proteins in GC cells,indicating that its cancerpromoting effect may be related to EMT process.
