ObjectiveThyroid cancer is a tumor that originates from thyroid cells.It is the most common endocrine tumor and one of the tumors with good prognosis.But in recent years,the incidence rate of thyroid cancer has been rising,especially in children and adolescents.ZBTB protein has a C-terminal zinc finger and N-terminal BTB/POZ domain,which is a new family of transcription factors.The ZBTB family participates in the occurrence of various biological processes,can regulate the development of specific lineages in the immune system,and plays a key role in development,differentiation,and tumorigenesis.Some genes in the ZBTB family are associated with the occurrence and development of thyroid cancer,but the expression and prognostic value of ZBTB in thyroid cancer are unclear.Therefore,this study aims to analyze the expression of ZBTB protein family in differentiated thyroid cancer,and explore its relationship with pathology and immune cells in differentiated thyroid cancer,so as to clarify the relationship between ZBTB family and the occurrence and development of differentiated thyroid cancer.Methods(1)TCGA database and R v4.0.3 software package were used to analyze the differential expression of ZBTB protein family in differentiated thyroid cancer and the correlation between genes.(2)The GEPIA2 database analyzed the relationship between differentially expressed genes and pathological staging of thyroid cancer.(3)The cluster profile v4 2.0 and org Hs.eg.db v3.14.0 were used for GO and KEGG enrichment analysis.(4)Kaplan Meier Plotter platform finds genes with differential expression of ZBTB protein family and prognostic value in thyroid cancer.(5)The TIMER database analyzed the correlation between prognostic genes and immune infiltrating cells.(6)STRING and In Bio Discover databases were used to construct protein network interaction diagrams for BCL6 B.(7)RT-q PCR was used to quantitatively analyze the expression of prognostic genes in differentiated thyroid cancer and normal tissues.(8)After knocking down the expression of the target gene through SiRNA,CCK-8 was performed to detect the proliferation of differentiated thyroid cancer cells;The scratch test detects the migration ability of cancer cells and the apoptosis detection kit detects the apoptosis value of cancer cells.Results(1)Bioinformatics analysis showed that compared to normal tissues,25 genes in the ZBTB protein family were down-regulated and 8 genes were up-regulated in differentiated thyroid cancer.In addition,survival analysis showed that low expression of ZBTB3,ZBTB9,ZBTB22,ZBTB34,ZBTB39,and high expression of BCL6 B had a worse prognosis in patients with differentiated thyroid cancer.(2)Through gene enrichment analysis,it was found that the ZBTB protein family is mainly involved in regulating hematopoiesis,regulating leukocyte differentiation,DNA binding transcription,and cancer transcription.(3)Prognostic genes ZBTB3,ZBTB9,ZBTB22,ZBTB34,ZBTB39,and BCL6 B were negatively correlated with CD8+T cells.(4)RT-q PCR analysis of thyroid cancer and normal tissue confirmed that BCL6 B was highly expressed in differentiated thyroid cancer.(5)Experiments on thyroid cancer cells have shown that knockdown of BCL6 B inhibits the proliferation and migration of thyroid cancer cells,and promotes apoptosis of differentiated thyroid cancer cells.Conclusions1.ZBTB3,ZBTB9,ZBTB22,ZBTB34,ZBTB39,and BCL6 B can be used as prognostic markers of differentiated thyroid cancer.2.Knocking down BCL6 B can affect the growth and migration of differentiated thyroid cancer cells.
