| ObjectiveThe purpose of this study was to investigate the expressions of FAM83A(Family with sequence similarity 83 member A)andβ-catenin in cervical adenocarcinoma,to analyze the relationship between the expression level and clinical pathology,and to explore the correlation between the expression of FAM83A andβ-catenin in cervical adenocarcinoma.MethodsGene expression profile Interactive Analysis 2.0(Gene expression profiling interactive analysis2.0,GEPIA2.0)database was used to analyze the expression of FAM83A gene in cervical cancer tissues and draw the survival curve.The overall survival time was analyzed.UALCAN database was used to analyze the relationship between abnormal expression of FAM83A and clinicopathological features.The FAM83A co-expression genes were identified by Linked Omics database,and then the co-expression genes were analyzed by gene ontology(Gene Ontology,GO)and genome encyclopedia(Kyoto encyclopedia of genesand genomes,KEGG)by R language(R package"cluster Profiler").Due to the small number of clinical cases of cervical adenocarcinoma,there is no independent distinction between cervical squamous cell carcinoma and cervical adenocarcinoma in the current public database,so this paper takes cervical cancer(including cervical squamous cell carcinoma and cervical adenocarcinoma)as the key words to analyze in the database.In addition,β-catenin is expressed in both normal epithelial cells and cancer cells.The difference is that in normal epithelial cells,β-catenin is only expressed in the cell membrane as an adhesion molecule,while in cancer cells,β-catenin is expressed ectopic in the nucleus or cytoplasm,and the two expression positions are different,so it is of no practical significance to analyze theβ-catenin coding gene CTNNB1 in online database analysis.CTNNB1 is not analyzed in this paper.Paraffin tissue specimens of cervical adenocarcinoma and non-diseased cervix were collected from the Department of Pathology of Shenyang Maternal and Infant Hospital from January 2011 to December 2021,including 89 cases of cervical adenocarcinoma and 86 cases of non-diseased cervix.Immunohistochemical method was used to detect the expression of FAM83A andβ-catenin in nondiseased cervical tissues and cervical adenocarcinoma,to explore the relationship between the expression level of FAM83A andβ-catenin in cervical adenocarcinoma and the clinicopathological features of patients,and to analyze the correlation between FAM83A andβ-catenin.Results1.According to the analysis of GEPIA2.0 database,it was found that the expression of FAM83A was abnormally high in cervical cancer,and the patients with high expression of FAM83A in cervical cancer had a poor prognosis.Using UALCAN database analysis,the results showed that the expression level of FAM83A in cervical cancer was related to the clinical stage(stage Ⅰ,Ⅱ,Ⅲ,Ⅳ).The later the stage,the more significant the expression of FAM83A,but there was no significant difference in the expression level of FAM83A among patients with different age and body weight.GO and KEGG enrichment analysis using Linked Omics database showed that there was a positive correlation between FAM83A and Wnt/β-catenin signal pathway.2.The results of immunohistochemistry showed that the expression rate of FAM83A in cervical adenocarcinoma was significantly higher than that in non-tumor cervical tissue(P<0.001),and the expression rate ofβ-catenin in cervical adenocarcinoma was much higher than that in non-tumor cervical tissue(P<0.001).3.The expression of FAM83A was not correlated with the age of the patients(P=0.344),but correlated with the differentiation degree of cervical adenocarcinoma(χ~2=9.021,P=0.006),tumor size(χ~2=25.610,P<0.001))and clinical stage(χ~2=57.099,P<0.001).The expression ofβ-catenin was not related to the age of the patients(P=0.466),but related to the differentiation degree of cervical adenocarcinoma(χ~2=10.786,P=0.001),tumor size(χ~2=14.990,P<0.001)and clinical stage(χ~2=27.606,P<0.001).4.There was a positive correlation between the expression of FAM83A andβ-catenin in cervical adenocarcinoma(r=0.706,P<0.001).Conclusions1.FAM83A is highly expressed in cervical cancer and is related to poor prognosis.There is a positive correlation between FAM83A and Wnt/β-catenin signal pathway.2.Both FAM83A andβ-catenin are highly expressed in cervical adenocarcinoma,which may be potential carcinogenic molecules in the development of cervical adenocarcinoma.3.The expression of FAM83A andβ-catenin was related to the differentiation degree,tumor size and clinical stage of cervical adenocarcinoma,especially in the tissues with low differentiation and late clinical stage,but FAM83A andβ-catenin were not related to the age of the patients.4.There is a positive correlation between the expression of FAM83A andβ-catenin. |
