| Objective:In order to explore the core prescription for the treatment of deep venousthrombosis,data mining was used to analyze and collect the cases of treating deep venousthrombosis(damp-heat type)in Jinan Hospital of Traditional Chinese Medicine.To offer ideas and basis for the clinical treatment of deep venousthrombosis,and to explore the effect and mechanism of Yinchen in the treatment of deep venousthrombosis through animal experiments under the guidance of network pharmacology.Methods:1.Clinical study: TCM prescriptions of patients with deep venousthrombosis(damp-heat type)admitted to the surgical outpatient department of our hospital from November 2020 to November 2022 were selected,sorted out and screened,and a TCM prescription database was established.The frequency of drug use,normalization flavor,association rules and complex network were analyzed by Microsoft Excel 365,SPSS Modeler 18.0 and Cytoscape 3.10.0.2.Animal experiment: Under the guidance of modern TCM research ideas on pharmacokinetic process and network pharmacology in vivo,our research group has found that the active component of Yinchen may play a role in the treatment of DVT by mediating NF-κB/MAPK/PI3K-Akt and other signaling pathways.Based on this study,60 SD rats were randomly divided into black group,model group and Yinchen intervention group.The blank group and the model group were given normal saline by gavage every day.The Yinchen intervention group was given Yinchen concentrate by gavage once a day for 7 days.After completing sample collection,the expressions and contents of NF-κB p65,IκBα,PI3 K,AKT,MAPK p38 and ERK proteins were detected by immunohistochemistry and Western Blot.Results:1.Data mining results: A total of 150 TCM prescriptions were collected,including 54 TCM flavors,among which 20 TCM flavors were used more than or equal to 30 times.According to the frequency of use,they were Yinchen,Coix seed,Phellodendron chinensis,Radix sophora flavescens,atractylodes atractylodes,Radix paeoniae,Danshen,licorice,alisma alisma,ligusticum ligusticum,Radix xylorhizae,Radix angelicae,honeysuckle,poria cocos,Radix usticum Rhizome and Radix xylorhizae radix.Among them,the herbs with the highest frequency of use were those that benefit water and permeate moisture,and the four herbs were mainly cold drugs,followed by warm drugs.The five tastes are mainly bitter,followed by sweet;Liver channel is the main channel,followed by spleen channel.The commonly used combinations of traditional Chinese medicine include Coix seed-Yinchen,Fang Ji-Coix seed,sophora flavescens-Phellodendron,etc.The core prescription of complex network is composed of Yinchen,Coix seed,Phellodendron chinensis,sophora flavescentis,atractylodes atractylodes and tetrandrhexis.2.Animal experiment results(1)The expression of NF-κB p65,IκBα,PI3 K,AKT,MAPK p38 and ERK was determinedImmunohistochemical staining was performed to observe the expression of NF-κB p65,IκBα,PI3 K,AKT,MAPK p38 and ERK.Compared with the blank group,the positive expression of NF-κB p65,PI3 K,AKT,MAPK p38 and ERK in the model group and Yinchen intervention group increased.The positive expression of IκBα decreased,(P<0.05).Compared with the model group,the expression of NF-κB p65,PI3 K,AKT,MAPK p38 and ERK in the Yinchen intervention group decreased,and the expression of IκBαincreased,the differences were statistically significant(P< 0.05).Western Blot showed that compared with the other two groups,the protein expression levels of NF-κB p65,PI3 K,AKT,MAPK p38,and ERK in the model group were the highest,and the protein expression level of IκBα was the lowest(P<0.05).Compared with the blank group,the protein expression of NF-κB p65,PI3 K,AKT,MAPK p38 and ERK in the Yinchen intervention group increased,and the protein expression of IκBα decreased,(P<0.05).Compared with the model group,the expression of NF-κB p65,PI3 K,AKT,MAPK p38 and ERK protein in the intervention group decreased,and the expression of IκBα increased,the differences were statistically significant(P< 0.05).Conclusion:Through data mining,the core prescription of Jinan Traditional Chinese Medicine Hospital for the treatment of deep venousthrombosis(damp-heat downward injection type)was summarized as Yinchen-Chixiaodou decoction;Yinchen can reduce the inflammatory response of deep venousthrombosis in rats,and its clinical efficacy may be related to the regulation of NF-κB/MAPK/PI3K-Akt signaling pathway. |
