| Purpose:Collect the data of febrile diseases in the ’ Lin Zheng Zhi Nan Yi An’,classify and sort the drugs,excavate the law of drug use,further explore the main active components of drugs for the treatment of febrile diseases,and speculate their potential mechanism of action,in order to provide reference for clinical drug use.Material and method:The medical records of ’ Lin Zheng Zhi Nan Yi An’ were recorded,and the traditional Chinese medicine contained in the medical records were classified and summarized.Analyze the frequency of drug use,and count the sexual flavor meridian and efficacy of drugs.SPSS software was used for association rule statistics and cluster analysis of drugs to mine core drugs,drug pairs and new prescriptions.TCMSP and Uniprot database were used to analyze the active components and targets of drugs for treating febrile diseases.Cytoscape software was used to visualize the complex interaction relationship of drugs-active components-target proteins.Metascape database was used to analyze the interaction relationship and related pathways.Results:1.Frequency of medication: A total of 60 medical records with 102 herbs were included.The total frequency of medication was 427 times,and a total of 33 herbs with frequency ≥ 5.According to the descending order of the frequency of drug use,the top ten were Dihuang,Gancao,Maidong,Renshen,Xijiao,Ejiao,Xuanshen,Jingmi,Shigao,Zhusha2.Category of drug efficacy: 16 types of drugs were involved in 60 medical cases.Among102 drugs,29 were heat-clearing drugs,mainly heat-clearing and fire-clearing drugs.Heatclearing drugs were used up to 139 times,accounting for 32.55 % of the total frequency,followed by tonifying drugs,resuscitation drugs and sedative drugs.3.Sexual flavor meridian and efficacy of drugs: According to the Pharmacopoeia of the People’ s Republic of China and Dictionary of Traditional Chinese Pharmacy,the Sexualflavor meridian and efficacy of 102 drugs was statistically analyzed.Among them,cold drugs were the most,with a total of 49 flavors,followed by warm and flat drugs;the flavors were mainly sweet,with a total of 53 flavors.The descending order was bitter and pungent.In the60 cases,there were 50 herbs belonging to lung meridian,heart meridian and stomach meridian ranked second and third respectively.4.Analysis of medication association rules: according to the minimum confidence of 15%,the minimum support of 75% and the maximum number of previous items of 2,a total of 18 association rules were obtained.The confidence of Ejiao→Dihuang,Ejiao and Maidong →Dihuang is 100%.Ejiao → Maidong,Ejiao and Dihuang → Maidong,Shigao → Gancao,Zhusha → Shexiang,Zhusha→ Xijiao are also common compatibility combinations.5.Cluster analysis of drugs : Five groups of new drug cluster groups were obtained by cluster analysis of traditional Chinese medicine with frequency ≥ 5 times.C1 : Zhusha,Shexiang,Jinbo,Niuhuang,Renshen;C2 : Xijiao,Xuanshen,Shichangpu;C3 : Yujin,Zhuyexin;C4 :Gancao,Dazao,Dihuang,Ejiao,Maidong;C5 : Jingmi,Shigao,Zhuye,Zhimu,Banxia6.Drug active ingredients and targets : All active ingredients in the new prescription were screened according to OB ≥ 30 % and DL ≥ 0.18.A total of 146 effective components were obtained after deduplication,and quercetin,kaempferol,β-sitosterol,stigmasterol and(S)-naringenin were the core components.There are 255 corresponding gene targets after duplicate removal,and PTGS2,HSP90,CALM,ESR1,PPARG and AR were relatively important targets.7.Potential mechanism of drugs : GO enrichment analysis showed that the active components of core drugs mainly involved molecular functions such as nuclear receptor activity and transcription factor binding.GO-CC analysis showed that the potential targets were mainly located in the membrane raft,postsynaptic membrane,dendrite and other positions.A total of217 pathways were obtained by KEGG enrichment analysis,mainly involving pathways in cancer,lipid and atherosclerosis,Kaposi sarcoma-associated herpesvirus infection,humancytomegalovirus infection.Conclusion:1.In the treatment of febrile diseases,Ye Tianshi reused heat clearing drugs based on the principle of dispelling warm pathogen and taking care of Yin,at the same time,he applied tonifying drugs,giving consideration to eliminating evils to support healthy energy.2.According to the cluster analysis of high-frequency drugs,the core drugs of the new prescription are Zhusha,Shexiang,Jinbo,Niuhuang,Renshen,Xijiao,Xuanshen,Shichangpu,Yujin,Zhuyexin,Gancao,Dazao,Dihuang,Ejiao,Maidong,Jingmi,Shigao,Zhuye,Zhimu,Banxia.3.The core drugs of the new prescription have the characteristics of multi-component,multitarget and multi-pathway.The main mechanism may be related to the reduction of inflammatory response,and it is speculated that the core components of the new prescription are mainly regulated through PI3K/AKT signaling pathway and MAPK signaling pathway. |
