Objective:To observe the effect of Mongolian medicine Hei Suga-25 on the behavior,morphology and the expression of nerve growth factor(NGF)and neurotrophin-3(NT-3)in hippocampus CA1 of vascular dementia model rats,and to explore the effective way of Hei Suga-25 in the treatment of VD.Methods:70 healthy SD rats(weight 140-180g)were randomly divided into 7 groups,including normal group,sham operation group,model group,positive control group,low dose group,medium dose group and high dose group,with 10 rats in each group.In the sham operation group,both common carotid arteries were exposed by skin incision and then sutured.The VD rat model was prepared by permanent ligation of both common carotid arteries in positive control group,low dose group,medium dose group and high dose group.The positive control group was given Donepezil 0.5mg/kg suspension,once a day for 28 days;The low dose group was given Hei Suga-25 at 90mg/kg,the medium dose group was given Hei Suga-25 at 180mg/kg,and the high dose group was given Hei Suga-25 at 360mg/kg,and the suspension was prepared by gavage once a day for 28 days.Morris water maze test and fatigue rotary rod test were used to detect the learning and memory function of rats in each group.HE staining was used to observe the pathological changes in hippocampal CA1 region of rats in each group.Immunohistochemical staining was used to observe the expressions of NGF and NT-3 in hippocampal CA1 region of rats in each group.Result:1.Morris water maze testThere was no significant difference between the sham group and the normal group(P>0.05),the model group had statistical significance(P< 0.05),indicating that cognitive function was impaired in model group;Compared with the model group,there was statistical significance in each administration group(P<0.05),indicating that the learning and memory ability of rats was improved after treatment;Compared with positive control group,there was no statistical significance in Hei Suga-25 high-dose group(P>0.05),Hei Suga-25 low dose group and Hei Suga-25 medium dose group had statistical significance(P<0.05);There was no significant difference between medium dose group and low dose group(P>0.05).2.Fatigue rotating rod test There was no statistical significance in the retention time of the rotating rod between the normal group and the sham group(P>0.05),compared with normal group,the retention time of rats in model group was significantly shortened(P<0.05).Compared with model group,stick retention time of rats in administration groups was significantly prolonged(P<0.05);Compared with positive control group,there was no statistical significance in the retention time on the rod in Hei Suga-25 high-dose group(P>0.05),Hei Suga-25 low-dose group and Hei Suga-25 medium-dose group had relatively shorter residence time(P< 0.05);There was no significant difference between low dose and medium dose groups(P>0.05).3.Results of HE staining in CA1 region of rat hippocampus:Under light microscope,the cells in the hippocampal CA1 region of rats in the normal group and the sham operation group were well organized,tightly arranged,and the cell morphology was intact,and no obvious pathological changes were observed.In model group,the cells in hippocampal CA1 region were loose,irregular in shape,blurred or disappeared in structure,and poorly arranged.Compared with model group,the morphology of CA1 cells in hippocampal CA1 region of rats in all administration groups was orderly,with clear boundaries,clear nucleoli,abundant cytoplasm and uniform coloring.4.Immunohistochemistry was used to detect the expression of NGF and NT-3integrated optical density in hippocampal CA1 region of rats:Compared with normal group,there were no significant differences in NGF and NT-3integral optical density of hippocampus in sham operation group(P>0.05),the integrated optical density in hippocampal CA1 region decreased in model group(P<0.05);Compared with model group,the integrated optical density of NGF and NT-3 in hippocampal CA1 region of rats in each administration group increased,with statistical significance(P< 0.05).Conclusion:(1)The learning and memory ability of VD model rats was improved after treatment with Hei Suga-25.(2)The morphology of nerve cells in hippocampal CA1 region of VD model rats was improved after treatment with Hei Suga-25,which showed protective effect on nerve cells.(3)Hei Suga-25 protects neurons by increasing the expression of NGF and NT-3 in hippocampal CA1 region of rats,thereby improving the cognitive function of VD rats.
