Background and Objectives:Autologous adipose tissue has been widely used in plastic surgery and plastic surgery due to its easy availability,abundant and cheap.In autologous fat transplantation,centrifugation based on gradient density sorting can remove excess oil and swelling fluid in adipose tissue.In previous studies,adipose tissue after centrifugation will form a density gradient.Compared with low-density fat,highdensity fat has more stem cell content and a higher retention rate after transplantation.Although some scholars use the three-part method to transplant the fat after centrifugation in layers,in clinical practice,the fat tissues of different individuals have different levels of oil and extracellular matrix.The fat of different individuals is treated by this method.The transplant retention effect is also different.Therefore,our previous research has found a more accurate method to screen the parts of adipose tissue that are more suitable for transplantation in different individuals,that is,by adding a density of 0.935g/ml to the Coleman centrifugation method.The standard density float divides the centrifuged fat into high-quality fat(HQF)and lowquality fat.However,we found that different individuals get different proportions of highquality fat.Therefore,this experiment conducted fat characteristics and animal experiments on the high-quality fat obtained by different individuals,and established the relationship between individual body data and high-quality fat,aiming to explore whether the high-quality fat obtained by different individuals has similar characteristics and graft retention rate.And the relationship between individual body data and high-quality fat,in order to improve the predictability of fat transplantation.Methods and materials1.Research on the characteristics of high-quality fat in different individuals and the outcome of graftsThe lipoaspirate was processed by Coleman centrifugation(1200g,3min)with a density of 0.935g/ml floater(1200g,3min),and HQF of different individuals was collected.The SVF cell count and ECM content percentage of different individuals HQF were determined to evaluate the characteristics of different individuals HQF.In animal experiments,different individuals were divided into lean,superrecombinant,and obese groups.The obtained HQF was transplanted into nude mice,and Coleman fat was used as the transplantation control group.The graft retention rate and graft structure at different time points were compared to explore the acquisition of different individuals.Does HQF have similar graft retention rates.2.Explore the relationship between high-quality fat of different individuals and individual body dataMeasure and collect relevant body data of healthy women who have received abdominal liposuction,use SPSS software for statistical analysis of data,and establish a linear regression relationship between individual clinical data and the percentage of HQF to analyze the correlation and build a model.Results:1.The results of in vitro experiments suggest that HQF obtained by different individuals have similar SVF cell numbers and ECM content.In animal experiments,at different time points,the appearance,retention rate,HE staining and immunohistochemistry results of HQF grafts were similar,but Coleman fat was different.2.Statistical analysis shows that the percentage of HQF has the greatest correlation with the body fat ratio of an individual.The higher the body fat ratio,the lower the HQF percentage.That is,in individuals with higher body fat ratios,the same volume of fat aspirate can be obtained have less HQF after processing than individuals with lower body fat ratio.ConclusionThe high-quality fat obtained by different individuals has similar cytological and histological characteristics,and similar transplantation processes and outcomes.The percentage of high-quality fat obtained by different individuals has the greatest relationship with the individual’s body fat ratio and is negatively correlated.The required fat suction amount can be roughly estimated through the model formula,which improves the predictability of fat transplantation.
