BackroundSub-health status is a status between health and disease.It can develop into a disease state after being in a sub-health state for a long time.At present,it is considered that sub-health status is the result of multiple factors,but its mechanism is not clear.More and more evidence reveals that intestinal flora is closely related to sub-health status.However,due to the different research schemes and gene sequencing technology,there is no unified conclusion on the changes and characteristics of intestinal flora in population with sub-health status.Objectives(1)To understand the current situation of night shift nurses with sub-health status and explore the related factors,so as to provide basis for their health management.(2)To study the change characteristics of intestinal flora of night shift nurses with sub-health status,so as to provide new ideas for the prevention and treatment of sub-health status.Methods(1)With convenience sampling,425 night shift nurses from Nanfang Hospital of Southern Medical University from November 2020 to may 2021 were selected as the research objects and surveyed by using Sub-Health Measurement Scale Version 1.0.Multivariable logistic regression was used to analyze the risk factors of sub-health status of night shift nurses.(2)Using the convenient sampling method,75 night shift nurses with sub-health status who met the inclusion criteria were selected as the sub-health status group(group SHS),and 75 healthy night shift nurses matched with gender,age and BMI were selected as the control group(Group H).Two groups of fecal samples were collected,and the V3-V4 region of bacterial 16S rRNA was sequenced and bioinformatics analyzed by High-throughput sequencing.Results(1)The incidence of sub-health of 425 night shift nurses was 77.6%,and the total average score of SHMS V1.0 was(52.06 ± 10.28),and the score of physical symptoms was the lowest.Multivariable logistic regression analysis showed that weekly exercise frequency,weekly meal frequency,daily average sleep time and average interval of night shift were independent influencing factors of sub-health status of night shift nurses(P<0.05).(2)The a diversity analysis showed that there was no significant difference in Chao1 index between group SHS and group H(P=0.619),and the Shannon index in group SHS was increased,and there was significant difference between the two groups(P<0.001),the β diversity analysis showed that there was significant difference in community structure between group SHS and group H(R=0.227,P=0.001);Species differences analysis showed that there were significant differences in relative abundance between the two groups in 4 phyla,3 classes,3 orders,3 families and 3 genera(P<0.05).Conclusions(1)he incidence of sub-health of 425 night shift nurses was 77.6%,and the score of physical symptoms was the lowest.The risk factors of sub-health status of night shift nurses are weekly exercise frequency,weekly meal frequency,daily average sleep time and average interval of night shift.(2)Compared with the healthy night shift nurses,the intestinal flora structure of night shift nurses in sub-health status is obviously unbalanced,and the ratio of Firmicutes to Bacteroidetes is increased,which may be related to the occurrence and development of sub-health status.We further defined the different species between the two groups,in which Blautia and Agathobacter in population with sub-health status increased significantly and Prevotella decreased significantly,but the specific mechanism needs to be further studied and confirmed.
