Effect Of Qinbaiqingfei Concentrated Pill And Its Active Ingredient Baicalin On Expression Of Cough Factor TRPA1

Objective:Illustrated in this paper,the effects of baicalin and its active components on the expression of cough factors TRPA1,CGRP and SP in Qinbaiqingfei Concentrated pills were discussed from animal experiments and molecular levels.The inhibitory effect of baicalin,the active ingredient of Baicalbaicalin,on mycoplasma pneumoniae pneumonia cough was analyzed and expounded,which provided theoretical and experimental basis for its further development,and could be used as the basis of clinical drugs supplement for the treatment of mycoplasma pneumoniae pneumonia cough.Methods:Biacore T200 fishing method was used to enrich,screen and recover the affinity active components of Qinbaiqingfei concentrated pills,and upl C-Q-TOF/MS was used to analyze and identify the recovered samples.Mycoplasma pneumoniae C57 mice were used to establish mycoplasma pneumoniae infection model.Lung tissues of mice were collected,and morphological changes of lung tissues were observed by HE staining.Changes in protein expression level and m RNA expression of cough factors TRPA1,CGRP and SP were detected by RT-PCR and immunohistochemical methods before and after administration of mycoplasma pneumoniae.Results:After Biacore T200 fishing analysis results showed that the active ingredient is baicalin as the active ingredient in Qinbaiqingfei concentrated pills,then through UPLC-Q-TOF-MS detection and analysis,verify the existing database,the results show that the active ingredient is indeed baicalin.HE staining showed that the lung morphology of baicalin group was significantly improved compared with that of model group.Real-time PCR results showed that compared with the model group,the m RNA expressions of TRPA1,CGRP and SP in baicalin administration group were decreased.Compared with the model group,the protein expression levels of TRPA1,CGRP and SP in baicalin group were significantly decreased.Conclusion:In this study,baicalin,the active ingredient in Qinbaiqingfei Concentrated pills,was extracted and identified by Biacore T-200.Related pharmacological experiments also verified that baicalin could inhibit the expression of cough factors TRPA1,CGRP and SP.This experiment showed that Qin best effective active ingredients baicalin in clearing lung concentrated pill can effectively inhibit cough after pneumonia mycoplasma infection,reveal Qin hundreds of clearing lung concentrated pill can,through its active ingredients baicalin of traditional Chinese medicine in the treatment of mycoplasma pneumonia pharmacodynamic material basis and mechanism of promoting drug research and development,the scientific significance and application prospect.
