| Objective:By observing the effect of electroacupuncture preconditioning on cardiac function,pathological changes of myocardial tissue and inflammatoryreaction in rats with isolated myocardial ischemia and reperfusion injury,to discuss the protective effect and mechanism of electroacupuncturepreconditioning on myocardial ischemia and reperfusion injury.Methods:In this experiment,forty-eight clean-grade SD male rats were selected and randomly divided into control group,model group,electroacupuncture Shenmen Xinshu preconditioning group(Shenmen Xinshu group),and electroacupuncture Taiyuan Feishu preconditioning group(Taiyuan Feishu group),and 12 rats in each group.A myocardial ischemia-reperfusion injury model was established using the Langendorff isolated reperfusion system.The isolated heart of the control group was balanced perfused with 180minutes.The model group,Shenmen Xinshu group and Taiyuan Feishu group were first balanced perfused with 20 minutes,then ischemic 40 minutes,and finally reperfused 120 minutes.The Shenmen Xinshu group and the Taiyuan Feishu group were treated with acupuncture once a day,each time 30 minutes,6 days a week for 2 weeks.Cardiac function was monitored during the experiment,pathological changes of myocardial tissue were observed by HE staining,by enzyme linked immunosorbent assay detected the content of TNF-αand IL-10,and the expression level of TLR4,NF-κBp65,TNF-αand IL-10 protein were detected by Western Blot method.Results:1.Cardiac function indexes in each group:compared with the control group,HR,LVDP,+dp/dtmaxand-dp/dtmaxdecreased and LVEDP increased in the other three groups(P<0.05).Compared with the model group and the Taiyuan Feishu group,HR,LVDP,+dp/dtmaxand-dp/dtmaxincreased and LVEDP decreased in the Shenmen Xinshu group(P<0.05).Compared with the model group,HR,LVDP,+dp/dtmaxand-dp/dtmaxincreased,and LVEDP decreased in the Taiyuan Feishu group(P>0.05).2.Pathological changes of myocardial tissue:in the control group,the myocardial tissue muscle fibers were arranged closely and neatly,without fracture,the boundary of nucleus and cytoplasm was clear,the structure was complete and stained completely,and there was no edema and inflammatory cell infiltration.In the model group,the arrangement of myocardial fibers was disordered,swollen,even broken,a large number of inflammatory cells infiltrated,the cell structure was destroyed and stained unevenly,and the areas of hemorrhage and necrosis could be seen.Compared with the model group,the Taiyuan Feishu group had slightly neat arrangement of myocardial tissue fibers,partial breakage,incomplete cardiomyocyte structure and more interstitial inflammatory cell infiltration.In the Shenmen Xinshu group,the myocardial fibers were arranged neatly,the edema was mild,the cell structure was complete,and there were still a few inflammatory cells in the stroma.3.The content of TNF-αand IL-10 in the myocardial tissue:compared with the control group,the content of TNF-αand IL-10 in the myocardial tissue of the model group increased(P<0.05).Compared with the model group and the Taiyuan Feishu group,the content of TNF-αdecreased and IL-10 increased in the Shenmen Xinshu group(P<0.05).Compared with the model group,the content of TNF-αdecreased and IL-10 increased in the Taiyuan Feishu group(P>0.05).4.Expression of TLR4,NF-κBp65,TNF-αand IL-10 protein inmyocardial tissue:compared with the control group,the expression level of TLR4,NF-κBp65,TNF-αand IL-10 protein in the myocardial tissue of model group increased(P<0.05).Compared with the model group and the Taiyuan Feishu group,the expression of TLR4,NF-κBp65 and TNF-αprotein in the myocardial tissue of the Shenmen Xinshu group decreased,while theexpression of IL-10 protein increased(P<0.05).Compared with the model group,the expression levels of TLR4,NF-κBp65 and TNF-αproteindecreased and IL-10 increased in the Taiyuan Feishu group(P>0.05).Conclusions:1.Electroacupuncture at Shenmen point and Xinshu point could improve the left ventricular function of rats with myocardial ischemia reperfusion injury,reduce the degree of pathological damage of myocardial tissue,and play a role in myocardial protection.2.Electroacupuncture at Shenmen point and Xinshu point may regulate the content of antiinflammatory factor IL-10 and proinflammatory factor TNF-αin myocardial tissue of rats with myocardial ischemia and reperfusion through TLR4/NF-κB pathway,so as to inhibit inflammatory response and protect the damaged myocardium.3.The intervention effect of electroacupuncture at Shenmen point and Xinshu point is significantly better than that of electroacupuncture at Taiyuan point and Feishu point,which has application value in the prevention and treatment of myocardial ischemia and reperfusion injury. |
