Objective: To explore the effective chemical active components and potential molecular mechanism of Yi Qi Qing Du Prevented Cancer Tea in preventing lung cancer by using the method of network pharmacology.Methods: The chemical constituents and drug targets of Yi Qi Qing Du Prevented Cancer Tea were queried through the database of Traditional Chinese Medicine System Pharmacology Database and Analysis Platform(TCMSP),and by setting the oral bioavailability(OB)≥ 40% and drug-like properties(DL)≥ 0.18 to select effective active ingredients and effective targets,and finally consult the literature to check and supplement targets through China National Knowledge Infrastructure(CNKI)database.The target results were checked and corrected in the Uniprot database,and finally the effective target information of Yi Qi Qing Du Prevented Cancer Tea was obtained.Based on the human gene database(Gene Card),with "lung cancer" as the key word,the species was set as Homosapiens to search for lung cancer related targets,and the Uniprot database was used to query the gene names corresponding to possible target proteins.Finally,standardizing the target protein.In the Venny online software mapping tool platform,the target genes of Yi Qi Qing Du Prevented Cancer Tea and lung cancer were respectively entered,and a Venn diagram was drawn to obtain the intersection of the two as potential targets.The screened potential target genes were imported into the STRING(Protein-Protein Interaction Network,PPIN)network platform,and the species was defined as Homosapis for analysis to obtain protein interaction information.The target was imported into Cytoscape 3.7.1 software to draw a PPI diagram(protein interaction network diagram),in which the change of the color depth and the size of the nodes both represented the magnitude of the degree value,and targets of core were screened out according to the size of the degree value of the nodes.The DAVID(DAVID Bioinformatics Resources)platform was used to perform enrichment analysis of potential target genes to obtain GO(Gene Ontology)and KEGG(Kyoto Encyclopedia of Genes and Genomes)enrichment analysis to predict the main targets of chemical constituents and the potential biological activity mechanism of Yi Qi Qing Du Prevented Cancer Tea,In order to explore the most likely mechanism of action of Yi Qi Qing Du Prevented Cancer Tea.Results: The results of network pharmacology research showed that 133 active ingredients were screened from Yi Qi Qing Du Prevented Cancer Tea,among which the active chemical ingredients were quercetin,kaempferol,catechin,isorhamnetin,mairin,jaranol,formononetin,calycosin,naringenin,stigmasterol etc.A total of 1244 effective targets were obtained,and 294 targets remained after the duplicates were deleted.23,648 lung cancer-related gene targets were obtained through the Gene Card database,and 390 gene targets with greater correlation were finally screened according to the median.Take the intersection of the two to obtain 91 drug-disease intersection targets,namely pharmacodynamic targets.535 biological processes(BP),50 cellular components(CC),and 87 molecular functions(MF)were obtained through DAVID database enrichment analysis of GO;KEGG enrichment analysis showed 117 signaling pathways.Among them,cancer pathway(hsa05200: Pathways in cancer),proteoglycans in cancer(hsa05205: Proteoglycans in cancer),hypoxia-inducible factor-1(HIF-1)signaling pathway(hsa04066: HIF-1 signaling pathway),tumor necrosis Factor(TNF)signaling pathway(hsa04668:TNF signaling pathway)are more important.Conclusion: The study of network pharmacology shows that Yi Qi Qing Du Prevented Cancer Tea has a preventive effect on lung cancer,and uses multiple active components such as quercetin,kaempferol,and catechin in various signaling pathways such as Pathways in cancer,Proteoglycans in cancer,HIF-1 signaling pathway,TNF signaling pathway,to act on EGFR,TP53,AKT1,PTEN and other genes can prevent the occurrence and development of lung cancer cells by inhibiting the cell cycle and angiogenesis in tissues.
