| Objective:To investigate the expression and correlation of miRNA-126 in Circulating monocytes with SPRED1,VEGF,ANG-1,ANG-2 in plasma of coronary heart disease.Methods:A total of 66 patients with coronary artery disease diagnosed by coronary angiography of People’s Hospital of Guangxi Zhuang Autonomous Region were enrolled,From December 2019 to December 2020.Which including 15 patients with acute myocardial infarction(AMI),26 patients with unstable angina(UA)and 25 patients with stable coronary heart disease(SCHD);17 cases were with single branch lesion,49 cases were with multiple branch lesion.Another 24 noncoronal heart disease people confirmed by coronary angiography were selected as the control group.At the same time extraction5ml venous blood was send to scientific experimental center to test.Circulating monocyte miRNA-126 levels were detected with qRT-PCR and SPRED1,VEGF,ANG-1,ANG-2 was detected by ELISA.Analyze the correlation between peripheral blood miRNA-126 level and SPRED1 in patients with coronary heart disease.Results:1.the expression level of miRNA-126 in coronary heart disease group was significantly higher than that in control group(P<0.05);2.The expression level of miRNA-126 in patients with acute myocardial infarction was higher than that in patients with stable coronary heart disease(P<0.05).The expression level of miRNA-126 in patients with unstable angina pectoris was higher than that in patients with stable coronary heart disease(P<0.05).There was no significant difference in miRNA-126 expression between patients with single vessel disease and patients with multi vessel disease(P>0.05);3.In AMI group,the expression level of SPRED1 was lower than that in SCHD and NCHD groups(P<0.05),the level of VEGF and ANG-2 was higher than that in UA,SCHD and NCHD groups(P<0.05),and the level of ANG-1was higher than that in NCHD group(P<0.05);4.The expression levels of miRNA-126 were highly negatively correlated with SPRED1(r_s=-0.829,P=0.000),highly positively correlated with VEGF(r_s=0.849,P=0.000),moderately positively correlated with ANG-1(r_s=0.496,P=0.06),but not significantly correlated with ANG-2(r_s=-0.154,P>0.1).Conclusion:1.The expression of miRNA-126 is up-regulated in peripheral blood mononuclear cells of patients with coronary heart disease.The expression level of miRNA-126 is significantly increased in patients with acute myocardial infarction and unstable angina pectoris.2.The expression of SPRED1 was down regulated in AMI patients,while VEGF,ANG-1 and ANG-2 were up-regulated.Mi RNA-126 was highly negatively correlated with SPRED1,suggesting that miRNA-126 may have a negative regulatory effect on SPRED1. |
