Effects Of MiR-145-5p On Biological Characteristics Of Bladder Cancer Cells Through The Regulation Of N-cadherin And ZEB2

Abstract/Summary:
Objective:Bladder cancer is one of the common tumors of the urinary system,with some characteristics of easy recurrence,metastasis and high mortality.The purpose of this study was to explore the relationship between miR-145-5p,ZEB2 and bladder cancer patients’ pathological data.Through overexpression/inhibition of miR145-5p and other methods,we investigated the effects of miR-145-5p on the proliferation,invasion,migration and other biological characteristics of T24,as well as the regulatory mechanism of miR-145-5p on ZEB2 and N-cadherin.Methods:A total of 34 postoperative cancer and paracancillary tissue samples from patients who suffered bladder cancer in the First Affiliated Hospital of Xiamen University from September 2018 to September 2020 were collected.The expression level of miR-145-5p was detected by qRT-PCR,and the detection data and clinical case data were statistically analyzed to clarify the relationship between miR-145-5p expression level and patient case data.Bladder cancer patients with ZEB2 expression data in TCGA database were screened,and the expression of ZEB2 and clinicopathological data were statistically analyzed to determine the relationship between the expression of ZEB2 and clinicopathological data.Through the transfection method,miR-145-5p was overexpressed or inhibited in bladder cancer cell line T24 to explore the effects of miR-145-5p on the biological characteristics of bladder cancer cells and to preliminarily explore the possible signaling pathways.Results:The expression of miR-145-5p in cancer and adjacent tissues was measured by qRT-PCR and the results have shown that the expression level of miR145-5p in bladder cancer tissues was lower than that in adjacent tissues.Furthermore,the expression level of miR-145-5p in invasive cancer tissues was dripped in noninvasive cancer tissues(P<0.05),and there was no significant difference with age,sex,tumor differentiation and tumor size(P>0.05).The expression level was negatively correlated with TNM grade and pathological stage of bladder cancer(P<0.05).In TCGA database,ZEB2 expression was positively in connection with TNM grade and pathological stage(P<0.05).There were no significant differences with gender,age,distant metastasis and survival time(P>0.05).Age,TNM stage,pathological grade and ZEB2 expression level were correlated with bladder cancer metastasis(P<0.05).After the transfection of miR-145-5p in bladder cancer cells T24,the expression of ZEB2 and N-cadherin weresignificantly down-regulated(P<0.05).CCK8 assay also showed that overexpression of miR-145-5p significantly suppressed the proliferation ability of T24 cells(P<0.05).In addition,the scratch test showed that the scratch healing area of T24 cells overexpressing miR-145-5p was smaller than that of the control group(P<0.05).Finally,Transwell assay showed that the membrane-penetrating ability of T24 cells which overexpressing miR-145-5p was reduced(P<0.05).Conclusion:Compared with adjacent tissues,miR-145-5p was lower expressed in BCa tissues,and the expression level was closely correlated with the clinicopathological data.Furthermore,the bladder cancer with high expression of miR145-5p tended to have better pathological staging.The expression level of ZEB2 in bladder cancer tissues was closely related to the clinical case data,and the high expression of ZEB2 in bladder cancer tissues tended to be in poor case stage.The overexpression of miR-145-5p in bladder cancer cell line T24 reduced the ability of proliferation,invasion and migration,and miR-145-5p could act on the downstream ZEB2 to affect N-cadherin,thereby inhibiting the EMT process of bladder cancer.
