| Bactrian camel,as a special animal group living in desert areas,has undergone some adaptive changes in its body tissues and organs under long-term natural evolution,and has a strong resistance to drought.The spleen is the largest peripheral immune lymphatic organ of the body,which can initiate the immune response,filter the aging and damaged red blood cells in the blood,and remove all kinds of antigenic substances in the blood.The spleen structure varies greatly among different animals,and even among the same animals,the spleen structure varies with age.So far,there are few studies on immune cells in Bactrian camel spleen.In order to clarify the biological characteristics of CD3εand CD4 in Bactrian camel and their distribution density in spleen with different ages,the basic physicochemical properties,functional sites and protein structures of CD3εand CD4 in Bactrian camel were analyzed by bioinformatics analysis software.At the same time,HE staining,PAS staining and Masson staining were used to analyze the size of splenic corpuscles,the distribution characteristics of glycoprotein,the distribution characteristics of spleen trabecular collagen fibers and cardiac muscle fibers of Bactrian camels at different ages.Immunohistochemical staining,immunofluorescence staining and statistical methods were used to analyze the CD3ε~+cells and CD4~+in the spleen of juvenile,young and adult Bactrian camels The distribution characteristics of CD4~+in cells were analyzed,and the results were as follows:(1)Morphological observation of the spleen in Bactrian camelThe splenic structure of Bactrian camels at different ages is basically the same,which is composed of a capsule and trabecula,marginal zone,white pulp and red pulp.The capsule is rich in elastic fibers and smooth muscle fibers,and the capsule connective tissue extends into the spleen to form trabecula.The white pulp is composed of periarterial lymphatic sheath and splenic nodules,which is equivalent to the cortex of lymph nodes.The red pulp consists of splenic cord and splenic blood sinuses.The statistical results of the size of splenic corpuscles in different age groups of Bactrian camels showed that the splenic nodule area of juvenile group,youth group and adult groups gradually increased,and the difference between juvenile group,youth group and adult group was significant.There was no significant difference between the youth group and the adult group.Distribution characteristics of glycoproteins:In all age groups,the glycoproteins in spleen were mainly distributed in the periarterial lymphatic sheath,marginal region and the red pulp,but less in the splenic nodules.Distribution characteristics of collagen fibers and smooth muscle fibers:spleen trabeculae in different age groups were mainly composed of collagen fibers,with a small amount of smooth muscle fibers.(2)Bioinformatics analysis of CD3εand CD4 molecules in Bactrian camelThe gene information of CD3εand CD4 molecules in Bactrian camel was searched on NCBI,and then transformed into amino acid sequence,and the bioinformatics analysis software was used to analyze.The results showed that CD3εis a stable transmembrane secreted protein.It is distributed in the extramembrane,cell membrane and cytoplasm,and its secondary structure is mainly random curling.CD4molecule is a relatively stable hydrophilic transmembrane secreted protein.The molecule is distributed in the extranembrane,cell membrane and cytoplasm,and has 5domains:one Ig domain,two Ig C2 domains,one CD4 extracellular domain,and one T-cell CD4 receptor C-terminal domain.The secondary structure of the molecule is dominated by random curling andβ-folding.(3)Expression characteristics of CD3ε~+cells and CD4~+cells in spleen of Bactrian camel at different agesImmunohistochemical and immunofluorescence results showed that CD3ε~+cells and CD4~+cells were distributed in the white pulp,the marginal area and the red pulp of the spleen,the distribution of the white pulp is more than the marginal area and the red pulp,the white pulp mainly distributed in the lymphatic sheath around the artery,the lymphatic vessels,the spleen nodules are also a small amount of distribution,the red pulp mainly distributed in the splenic cord.Statistical results showed that there was no significant difference in the distribution density of CD3ε~+cells in the white pulp between the juvenile group and the youth group,but the difference between the juvenile group and the adult group was very significant,and the difference between the youth group and the adult group was very significant.There was no significant difference in cell density between different age groups in the marginal zone and the red pulp.CD4 cells:in white pulp,there were very significant differences between juvenile group and youth group,juvenile group and adult group,and significant differences between youth group and adult group.In the marginal zone,there was no significant difference among the three age groups.In red pulp,there were significant differences between youth and adult groups,but no significant differences between other age groups. |
