| Plants act as seed dispersals for reproductive purposes through pods dehiscence,The MADS box family has been shown to be critical for organ formation during plant growth and development,Moreover,the functions of genes from this family,namely FUL and SHP,are associated with silique cracking.AGL8(AGAMOUS-LIKE 8)is also known as FUL.In this study,we used Caragana korshinskii and Arabidopsis thaliana to select gegens related to pod dehiscence and verify the biological function of the CkAGL8.The main results are as the following:1.The cDNA sequence of the CkAGL8 gene was cloned,in which the CDS was 738 bp long,encoded 245 aa;the protein molecular mass was 28291.31 Da,and the isoelectric point was 8.94.CkAGL8 was predicted to be localized in the nucleus.2.CkAGL8 was genetically transformed into A.thaliana.It was found that the valve margins and the replum of the silique dehiscence zone of the CkAGL8 overexpressed siliques tended to inosculate as a whole,which caused the structure of the dehiscence zone to change dramatically.Shatter resistant siliques was higher in the CkAGL8 transgenic line than that in wild-type,under the same temperature and the same time period treated.3.Transcriptome analysis of the valve margins from the dehiscenced pods and semi-dehiscenced pods of wild C.korshinskii identified a set of candidate genes associated with pod dehiscence.Further analysis of these genes will provide detailed insight into the mechanism of pod dehiscence of C.korshinskii.The above results indicate that heterologous expression of CkAGL8 in A.thaliana alter the architecture of the silique dehiscence zone,This gene causes overexpressed strains to exhibit pod shatter resistance. |
