Caragana intermedia Kuang distributes in the arid and semi-arid area,it is drought and extreme temperature tolerant,and was used widely in sand fixation,soil and water preservation with highecology value.And the it is an ideal material to study the resistance genes.NAC transcription factors had been proved to participate in plant response to various stresses.In this study,two NAC homo log genes.CiNAC036 and AtNAC036,were cloned by PCR amplification from C.intermedia and Arabidopsis thaliana respectively,and the transgenic Arabidopsis was subjected to drought,salt and mannitol stress to analyze the genes function.The results are as follows:1.An annotation of NAC gene sequence was obtained from the C.intermedia drought treated transcriptome database,and a comparison by Blast in NCBI showed that the gene had a complete open reading frame(ORF),and its protein sequence was 74%consistent with NAC036 in A.thaliana.Hence,the gene was named CiNAC036.2.The CiNAC036 was cloned and its cDNA is 909bp,encoding 302 amino acids,and its protein has a complete NAC domain.At the same time,AtNAC036 gene was cloned with the gene size is 831p,encoding 276 amino acids.3.After treatments by mannitol,drought and salt,only the mutant of AtNAC036 showed slightly sensitive under mannitol treatment,compared to the wild type;both the CiNAC036 and AtNAC036 transgenic Arabidosis did show obvious difference with wild type under all treatment conditions.4.CiNAC036 can promote the elongation of the main root of the transgenic Arabidopsis,while A tNAC036 seems to inhibit this process.Further study is still needed to explain the conflicting phenotype of these two homo log genes.
