In order to increase nitrogen absorption and reduce nitrogen loss of potato, the effects of Manure replacing nitrogen fertilizer on soil nitrogen pool and related microorganisms of potato farmland were clarified, and the effects of Manure replacing nitrogen fertilizer on dry matter quality and nitrogen absorption of potato during the whole growth period were studied. At the same time, soil mineral nitrogen content and soil nitrogen mineralization in different soil layers were measured, indicating the impact of Manure replacing nitrogen fertilizer on nitrogen supply of potato;Through the study of soil nitrogen forms and related microbial communities during potato tuber expansion, the effect of Manure replacing nitrogen fertilizer on soil nitrogen pool was explored. The main results are as follows:(1)Compared with the application of purified nitrogen fertilizer, the replacement of60% nitrogen fertilizer with Manure increased potato yield, nitrogen utilization efficiency and partial productivity of nitrogen fertilizer,as well as dry matter Cumulant and nitrogen Cumulant during potato tuber expansion.(2)Compared with the application of purified nitrogen fertilizer, the replacement of Manure with nitrogen fertilizer increased the content of soil mineral nitrogen and net mineralization of soil nitrogen during potato tuber expansion, and the replacement of 60%nitrogen fertilizer with Manure significantly reduced the apparent loss and surplus of nitrogen.(3)In potato tuber expansion stage, compared with the application of purified fertilizer nitrogen,Manure replaced 100% nitrogen fertilizer to increase soil microbial biomass nitrogen and soluble organic nitrogen. When Manure replaced 30% nitrogen fertilizer,it increased nitrate nitrogen and ammonium nitrogen content.(4)In the potato tuber expansion stage, compared with the nitrogen application of purified fertilizer, the substitution of Manure for nitrogen fertilizer increased the microbial diversity and richness. The dominant bacteria in the soil microbial community were Acidobacteriota,Proteobacteria,Gemmatimonadota,Bacteroidota and Actinobacteriota, respectively. The substitution of Manure for nitrogen fertilizer increased the abundance of Proteobacteria,Bacteroidota and Actinobacteriota, and reduced the abundance of Gemmatimonadota and Acidobacteriota. In addition, the change of nitrogen related microorganisms increased soil Nitrogen fixation related microorganisms and decreased nitrate reduction related microorganisms for Manure to replace nitrogen fertilizer. To sum up,the replacement of 60% nitrogen fertilizer with Manure can improve the potato yield by improving the nitrogen supply in the potato field and increasing the absorption of nitrogen nutrients in the potato, and at the same time change the process of transformation from organic nitrogen to mineral nitrogen, increase the diversity and richness of soil microorganisms, improve the microorganisms related to soil Nitrogen fixation, and reduce the loss of soil nitrogen.
