Abstract/Summary:
Immunopotentiator is a drug that can enhance the immune function of the body.Applying it as a feed additive to animal breeding can effectively reduce the disease rate of animals,thereby enhancing animal production capacity,and has certain positive significance for reducing the treatment cost in the breeding industry.At present,most of the research and application of immune enhancers in the breeding industry are concentrated on pigs and poultry industries with short growth cycles and high economic benefits.Domestic research and application of immune enhancers for ruminants account for very little.However,the actual effect of immunoglobulin preparations produced by a company in Zhejiang Province as an immune enhancer in mutton sheep breeding has not yet been clarified.Objective: In this experiment,by adding different doses of immunoglobulin preparations produced by a company in Zhejiang Province to the diet of experimental sheep,measuring the growth indicators and immune indicators of mutton sheep,and performing sequencing analysis on the rumen flora,so as to explore the product The effects on the growth performance,immune function,rumen flora and the optimal dosage of the test sheep provide a reference for its application as an immune enhancer in mutton sheep breeding.Methods: Thirty 90-day-old Kazakh fattening ewes were randomly divided into five groups: A,B,C,D,and E,respectively,the traditional Chinese medicine group,the 1% immunoglobulin preparation addition group,the 2% immunoglobulin preparation addition group,4% immunoglobulin preparation group,control group,6 sheep in each group.1.Determination of growth performance Weighing was carried out on test 1d,18 d,36d,54 d,and 139 d,and body measurements were carried out at the beginning and end of the test,so as to calculate the average daily weight gain,body length,body height,and chest circumference of the test sheep,The average growth of pipe girth.2.Determination of immune function On the basis of the time of the previous test,5 m L of blood was collected from the jugular vein of the test sheep and the serum was separated,and the IL-2 and IL-6.The content of Ig A,Ig G and Ig M,and the determination of secretory immunoglobulin.At the end of the test,3 sheep were randomly selected from each group of test sheep and slaughtered to take part of the duodenum,jejunum,and ileum,and collect mucous mucus.Use the sheep The content of s Ig A was detected by enzyme-linked immunosorbent assay kit.3.Determination of Small Intestine Morphology At the end of the experiment,the slaughtered sheep were taken from the duodenum,jejunum,and ileum to make intestinal tissue slices,and the villus height and crypt depth were measured and the villus crypt ratio was calculated.4.At the end of the experiment,the rumen fluid of 3 slaughtered sheep in each group was collected,sequenced,and the abundance and diversity of the flora were analyzed.Results: 1.There was no significant difference between the daily weight gain and the total weight gain of the mutton sheep in each immunoglobulin preparation group and the E group;the total growth of the chest circumference of the B group was the most significant(P<0.05);The growth rate was significantly higher than that of other groups(P<0.05).2.In terms of immune function,the levels of IL-2,Ig A,Ig G and Ig M in the serum of the test group were significantly higher than those of the E group(P<0.01),especially in the C group,and there was no significant difference in the level of IL-6 among the groups.The levels of s Ig A in groups B,C,and D were significantly higher than those in groups A and E(P<0.01),and the level of s Ig A in group C was the highest.3.In terms of small intestine morphology,the height of small intestinal villi and the depth of crypts in the experimental group were significantly different from those in the E group(P<0.01),especially in the D and A groups.A and B groups(P<0.01).4.The Shannon index of the three groups B,C,and D was slightly higher than that of the other groups but there was no significant difference(P>0.05),and the difference in the Simpson index of each group was not significant(P>0.05).All groups are dominated by Bacteroidetes and Firmicutes,and Bacteroidetes as the first dominant phylum occupies61.66% of the relative abundance in group C,which is higher than that in group E groups,but there was no significant difference in the abundance of flora among the groups(P>0.05).Conclusion: The immunoglobulin preparation produced by a company in Zhejiang Province can effectively enhance the immune function of mutton sheep,improve the mucosal immune function of the small intestine,and promote the development of small intestinal tissue.Among them,the immunoglobulin preparation with 2% additive dose in the diet has the best effect.
