Cancer is one of the most common diseases in dogs, especially with the increase of age, under the influence of internal and external environment, the incidence of cancer increases positively.Hospital generally after surgical resection of visible mass, continuous anti-cancer drugs, regular review observation; It is not possible to accurately judge the nature of the tumor and evaluate the prognosis of metastasis and recurrence of the tumor. At present, the pathological diagnosis basis of pet tumors in domestic clinical diagnosis and treatment is not perfect, and there is a lack of sufficient clinical data.In this study, dog tumor samples collected from animal hospitals in Urumqi and surrounding counties and cities were used as experimental objects.HE staining,PAS glycogen staining and Masson three-color staining were used for differential diagnosis and tumor types were determined. Thus, the similarities and differences between various tumor tissues can be found, and the relationship between tissues and tumorigenesis can be mastered to provide more abundant data for clinical diagnosis and treatment. Methods: The collected clinical tumor cases were analyzed with the relevant information such as variety, age, gender and site of disease.HE staining technique was used for pathological diagnosis and analysis of dog tumor sample sections to determine the type and nature of the tumor. At the same time,Masson three-color staining,PAS glycogen staining and immunohistochemical staining were combined for further differential diagnosis of canine tumors,so as to obtain accurate pathological diagnosis results, providing certain guiding significance for the clinical treatment and prognosis of pet physicians.Results:(1)Analysis of the characteristics of canine tumor;According to the analysis of 66 samples collected, the tissues with high incidence of tumor in dogs were breast tissue and skin tissue. The incidence of breast tumors was higher in female dogs than in male dogs. The incidence of breast tumors was higher in female dogs, while the incidence of other tumors showed no significant gender bias. The incidence of canine tumor begins in adulthood and peaks in adulthood and old age, suggesting that the incidence of canine tumor tends to be younger. The most affected breeds were teddy dogs, followed by Chinese pastoral dogs,Pomeranian dogs, etc.,which is closely related to people’s breeding environment and preferences.(2)Classification and characterization of tumors;Among the 66 clinical cases collected, there were 23 benign tumors and 43 malignant tumors.Among the benign tumors,12 cases were originated from skin tissue. The malignant tumors originated from breast tissue accounted for the largest proportion of 24 cases.(3)The application of special dyeing methods;Masson staining can distinguish stained fiber tissue, muscle fiber tissue and collagen fiber tissue, which is of great significance for the differential diagnosis of fiber tissue tumor and smooth muscle tumor.PAS glycogen staining can clearly express the presence or absence of glycogen in tissues, but in this study,it lacks differential diagnostic significance. The P53 tumor suppressor gene suggested histiocytosis, and carcinoembryonic antigen(CEA)suggested cell invasion and metastasis to surrounding tissues. Conclusions: A total of 66 tumor cases were collected from animal hospitals in Urumqi,Changji,Shihezi and Korla during 2022 to 2023.The study found the highest incidence of breast and skin tumors in dogs.High growth period in the prime and old age, adult and old age relatively less. The high prevalence breeds were teddy,Pomeranian, string and other small and medium-sized dogs. With the increase of dog size, the incidence decreased gradually.Among the 66 tumor cases involved in this study, there were only 23 benign tumors and 43 malignant tumors. The incidence of malignant tumors was significantly higher than that of benign tumors.Histopathological diagnosis is easy to diagnose tumor tissues with obvious features, but for tissues with complex components, various special staining methods are still needed to assist diagnosis.
