The genetic diversity and genetic relationship of peach and apricot varieties resources were reached by SSR markers,ITS and chloroplast sequences analysis three methods,preliminary obtained the following results:1.A collection of 95 peach cultivars,have been screened with 32 SSR primer pairs in order to analyze their genetic diversity.93 bands were amplified by 18 informative and reliable primers,of which 50 bands were polymorphic loci.Nei’s gene diversity 0.5652 and Shannons index 1.1125 was revealed at population level.The population genetic analysis based on peach maturity was classified early,middle and late maturity three groups..Late maturity group of Nei’s highest degree of genetic diversity and Shannon index,0.5879 and 1.1222 respectively;early maturity group of Nei’s lowest degree genetic diversity and genetic lowest phenotypic index,Shannon were 0.5301 and 1.0243.The genetic diversity and genetic heterozygosity of late maturity group > middle maturity group > early maturity group.The genetic consistency of early maturity groups and medium groups are closer,but late maturity are far away with both.Early and medium maturity groups had close relatives,the late groups had far relatives with the first two groups.The population genetic analysis based on peach shape was classified flat spherical and spherical two groups.Flat spherical group of Nei ’s degree of genetic diversity is higher than spherical group,flat spherical group had higher the genetic diversity.The population genetic analysis based on peach wild was classified wild and no wild two groups.,Wild group of Nei ’s degree of genetic diversity and Shannon index are higher than no wild group,the genetic diversity and genetic heterozygosity of wild group is higher than no wild group.2.A collection of 168 apricot cultivars,have been screened with 22 SSR primer pairs in order to analyze their genetic diversity.113 bands were amplified by 17 informative and reliable primers,of which 63 bands were polymorphic loci.Nei’s gene diversity 0.7158 and Shannons index 1.4973 was revealed at population level.The population genetic analysis based on apricot maturity was classified earliest,early,middle,late and latest maturity five groups.,Latest maturity group of Nei ’s highest degree of genetic diversity and Shannons index,0.7085 and 1.4416 respectively;earliest maturity group of Nei’s lowest degree genetic diversity and genetic lowest phenotypic index,Shannons were 0.6667 and 1.2736.Showed that the genetic diversity and genetic heterozygosity of latest maturity group is the highest,the genetic diversity and genetic heterozygosity of earliest maturity group is the lowest.The genetic consistency of early maturity groups and medium groups are closest,and earliest maturity are fariest with latest maturity group.This showed early and medium maturity groups had closest relatives,earliest and latest maturity groups had fariest relatives.The population genetic analysis based on apricot shape was classified oblatenes,circle,oval,ellipse,obround and heart-shape six groups.Obround group of Nei ’s highest degree of genetic diversity,0.7119;heart-shape group of Nei ’s highest degree of genetic diversity,0.6094,the genetic diversity of obround group is the highest,the genetic diversity of heart-shape group is the lowest.The genetic consistency of oval groups and circle groups are closest,is 0.9813,the genetic consistency of heart-shape group and ellipse group are farest,is0.7548.Showed showed oval group and circle group had closest relatives,heart-shape and ellipse group had fariest relatives.The population genetic analysis based on apricot core taste was classified sweet and bitter two groups.Sweet group of Nei ’s degree of genetic diversity and Shannons index are higher than bitter group,the genetic diversity and genetic heterozygosity of sweet taste group is higher than bitter group.3.Through the peach varieties and apricot varieties of all nine species for sequence alignment and ITS sequence clustering figure,it can be seen that is divided into two groups,the north and south each one big groups,which have peach and apricot,but can be separated in a large group of both.4.Through the peach varieties and apricot varieties chloroplast sequence psbA of nine varieties of sequence alignment and clustering figure,you can see Nanfang Jin Mi had no female parent relationship with the rest of the varieties.Ying Tiao had far relationship with the rest of the varieties of female parent.Closely female parent relationship between other peach varieties and Shuang Ren apricot,probably originated from the same female parent.Similarly,the rest of the apricot varieties may be originated from the same female parent... |
