The Esox lucius is one of the important characteristic fish in Xinjiang.Due to its unique biological value,In recent years,scholars at home and abroad have carried out a lot of research on genomics and gender.In order to further explore the molecular mechanism and gene network mechanism of gonadal differentiation in Esox lucius,In this study,the Esox lucius as the research object,collected the male and female gland tissues of Esox lucius at different developmental stages,sequenced the transcriptome,and made a preliminary analysis of the obtained data,and analyzed and screened the temporal differential expression of Esox lucius testis genes,ovarian temporally differentially expressed genes,and differentially expressed genes of male and female gland development,and GO enrichment analysis and KEGG pathway analysis were performed on the above differentially expressed genes.The experiment results show that:A total of 688960135 raw read pairs were measured in the transcriptome of male and female Esox lucius gonads.After quality control,672621722 clean read pairs were obtained by removing the linkers and low-quality data from the sequencing raw data.The total number of bases was 100.9Gb.Bases of more than 3.36 Gb were obtained,the average GC content of 30 samples was 48.12%,the Q20 was more than96.12%,and the Q30 was more than 90.26%.Through differential expression analysis,there were 15168 up-regulated genes and 15141down-regulated genes temporally differentially expressed in Esox lucius testis;9405 up-regulated genes and 8379 down-regulated genes in Esox lucius ovary;differences in male and female glands of Esox lucius There were 26858 up-regulated genes and 24263 down-regulated genes.Through the comparative analysis of differentially expressed genes in different periods,Through the comparative analysis of differentially expressed genes in different periods,it was inferred that the pike testis differentiated,and the ovarian development was concentrated at about 58 days after hatching.Through the annotation of GO and KEGG databases,the most abundant biological process GO terms include organic nitrogen compound metabolism,cellular protein metabolism,etc.;the most abundant cellular component GO terms include acetylcholine-gated channel complex;the most abundant molecular function GO Terms include anion binding,purine nucleotide binding,ribonucleotide binding,etc.The expression of genes related to the MAPK signaling pathway,endocytosis,neuroactive ligand-receptor interaction was more significant in KEGG database.The sequenced non-repetitive sequence genes(unigenes)were compared with the existing genome annotation information,and 5240 new transcripts were obtained.3179,4426,27,1412,2822996 and 634 new genes were successfully annotated in seven data including NR,NT,KO,Swiss Prot,Pfam,GO and COG/KOG,respectively.The results of this experiment provide the transcriptome information of male and female gonads in different growth and development stages of Esox lucius,lay a theoretical foundation for the regulation of gonad development and reproductive regulation in Esox lucius,and provide certain basic data for the study of the mechanism of sex differentiation in Esox lucius of Scientific value and theoretical significance.
