| Litopenaeus vannamei is one of the most popular seawater cultured species in China and even all over the world.But there is not a lot of basic research into it,and baseline information related to molecular regulatory mechanism of its gonad development is absent.In this study,we launched transcriptomic sequencing on 4 samples of the gonads of its broodstock,which were Testis,Ovary,1DaEAO(one Day afer Eyestalk Ablation Ovary)and 6DaEAO(6 Days afer Eyestalk Ablation Ovary),receiving 24,9107,536 raw reads.After quality control,filtrated and de novo assembled,65,218 Unigenes were generated from raw reads,with an average length of 1,021 bp and an N50 of 2,000 bp.Quantity and proportion of the Unigenes annotated to the NR library,NT library,Swiss-Prot library,KEGG library,COG library,GO library were 26,482(87.39%),14,626(48.26%),23,062(76.10%),20,659(68.17%),11,935(39.38%),12,320(40.65%).After the GO classification,there were 9,884 Unigenes in molecular function,while 19,939 Unigenes in cell components and 34,317 Unigenes in biological processes.The 20,659 Unigenes annotated to the KEGG library were mapped to 258 pathways,of which the top three were regulation of metabolic pathways,RNA transport and microfilament skeletons.By comparing the Ovary and the Testis library,there were 22,808 Unigenes showing differentially expression of which 3,529 were up-regulated in ovary and 19,279 were up-regulated in testis.After enrichment analysis,DEGs(differentially expressed genes)up-regulated in testis were classified into 414 enriched GO term while DEGs up-regulated in ovary were classified into 646,as well as being maped into 34 enriched KEGG pathways.After validating by quantitative real-time PCR,we identified 9 DEGs specifically expressing in ovary,6 DEGs specifically expressing in testis,39 DEGs up-regulated expressing in ovary,and 45 DEGs up-regulated expressing in testis.In addition,13,223 SSRs were found in 10,411 Unigenes,of which 2,192 Unigenes containing more than one SSR.This information will enrich molecular genetic information database and provide the basic information for the future study on the molecular mechanism and regulation of the gonadal development and the improvement of seed production technology and reproductive traits of Litopenaeus vannamei.SSRs can be used as molecular markers for for functional genomics research and developing a new molecular breeding technology. |
