By taking use of DNA sequencing and bioinformatics methods,this study mainly focused on50 Yili horses as research objects to study the polymorphism of MCT1、PPARα、NOS3、HIF-1αgenes in Yili horses,determine the genotypes of different mutation loci,and correlate the different genotypes with the athletic performance of Yili horses,so as to study the effect of MCT1,PPARα,NOS3 and HIF-1α genes on 2000 m athletic performance,exploring the mutation loci affecting the2000 m athletic performance of Yili horses,which provides a theoretical basis for early selection of speed horses.The study results are as below:(1)Based on the results analysis on sequencing of exons 2 and 3 of the MCT1 gene and the near region of the Yili horse,it showed: SNP1(g.52168582T>G)was detected in exon 2,but the mutation was not detected in exon 3.SNP1 was a missense mutation,of which three genotypes was detected in total,staying Hardy-Weinberg equilibrium and belonging to moderately polymorphic.From the analysis on the association between different genotypes of SNP1 in Yili horses’ MCT1 gene and 2000 m speed,it showed that the 2000 m speed of TT genotype in mutation loci g.52168582T>G was significantly faster than that of GG genotype(P<0.05).(2)Based on the results analysis on sequencing of exons 1,2 and 3 of the PPARα gene and the near region of the Yili horse,it showed: one mutation loci was respectively detected in exon 1and intron 3,SNP2(g.43225009T>G)and SNP3(g.43233663A>G),and the mutation loci was not detected in exons 2 and 3.SNP2 was an intronic mutation loci and SNP3 was an intronic mutation loci,in which all three genotypes were detected,staying in Hardy-Weinberg equilibrium and belonging to moderately polymorphic.From the analysis on the association between the different genotypes of SNP2 in Yili horses’ PPARα gene and 2000 m speed,it showed that the 2000 m speed of TT genotype in mutation loci g.43225009T>G was significantly faster than that of TG genotype(P<0.05).(3)Based on the results analysis on sequencing of exons 1 and 5-7 of the NOS3 gene and the near region of the Yili horse,it showed: SNP4(g.102724192G>A)and SNP5(g.102728344A>G)were respectively detected in intron 2 and exon 6.SNP4 was an intronic mutation loci and two genotypes were detected in total,which were low polymorphic loci;SNP5 is a missense mutation loci with three genotypes detected in total,which were moderately polymorphic loci.SNP4 and SNP5 were all staying in Hardy-Weinberg equilibrium.From the analysis on the association between the different genotypes of SNP5 in Yili horses’ NOS3 gene and 2000 m speed,it showed that the 2000 m speed of AA genotype in mutation loci g.102728344A>G was significantly faster than that of AG and GG genotype(P<0.05).(4)Based on the results analysis on sequencing of exons 3-4,7 and 9 of the HIF-1α gene and the near region of the Yili horse,it showed: only one mutation loci SNP6(g.8830772A>G)was detected in intron 2,therefore SNP6 is the intron mutation loci and two genotypes were detected in total,which were low polymorphic loci,staying in Hardy-Weinberg equilibrium.From the analysis on the association between the different genotypes of SNP6 in Yili horses’ HIF-1α gene and 2000 m speed,it showed that the 2000 m speed of AG genotype in mutation loci g.8830772A>G was faster than that of AA genotype,but the difference was not significant(P>0.05).To sum up,the Yili horses’ MCT1,PPARα,NOS3 and HIF-1α gene affect 2000 m athletic performance.These four genes are candidate genes affecting the athletic performance of speed horse.Enhancing the selection of these four genotypes in future specialized selection and breeding Improving the speed of Yili horse racing,improving the accuracy of Yili horse sport selection and early selection and breeding,accelerating the breeding process of specialized strain breeding of Yili horses,and having important strategic significance and practical application value for rapid selection and breeding of high quality sport Yili horse,improving the economic income of farmer and herdsmen,and accelerating the development of modern horse industry in Xinjiang.
