Association Of PFKM,PYGM,GYS And PRKAG3 Gene Expression With Glycolytic Potential In Different Tissues Of Yili Horse

Abstract/Summary:
The experiment selected eight Yili horses of similar age,physical condition and health from Ili Kazak Autonomous Prefecture.According to tissue samples were divided into Heart,Liver,Clamping,Trapezius,Latissimus Dorsi,Gluteals Medius,Semitendinosus and External Oblique 8 groups,The differences of glycolytic potential,hormones,enzymes and the m RNA expression of PFKM,PYGM,GYS and PRKAG3 genes among different tissues of horses were detected.Comprehensively analyzed the correlation between gene expression,glycolytic potential and enzymes in different tissues of Yili horse.The results showed that:Among the glycolysis indexes,the content of glycogen in semitendinosus,liver,musculus sandwich,trapezius,gluteus medius,latissimus dorsi,heart and external oblique vary significantly different(P<0.01).The content of glycogen in liver tissue was the highest,and the content of free glucose and total sugar in liver tissue was significantly higher than that in musculus sandwich,trapezius,latissimus dorsi,gluteus medius,semitendinosus.Under different glycolytic potential(GP),the glycogen concentration of high GP group vary significantly higher than that of low GP group(P<0.01),and the glycogen content of high GP group was significantly higher than that of low GP group(P<0.05);Under different glycolytic potential(GP),the concentration of lactic acid in the high GP group vary significantly higher than that in the low GP group(P<0.01),and the content of lactate in the high GP group was significantly higher than that in the low GP group(P<0.05),The glycolytic potential in liver tissue vary significantly higher than that in sandwich muscle,trapezius,latissimus dorsi,gluteus medius,semitendinosus,heart and external oblique(P<0.01).In the terms of enzyme activity,the content of hexokinase in heart tissue was significantly higher than that in sandwich muscle,trapezius,semitendinosus,latissimus dorsi,gluteus medius and external oblique muscle(P<0.01),and the content in liver tissue was significantly higher than that in heart tissue(P<0.01).The content of pyruvate kinase,phosphofructokinase and lactate dehydrogenase in liver was significantly higher than that in sandwich muscle,trapezius,latissimus dorsi.Under different glycolytic potential(GP),the content of pyruvate kinase in trapezius,latissimus dorsi and middle and high GP group was significantly higher than that in low GP group(P<0.01).Under the hormone indicators,the contents of insulin and epinephrine in liver tissue were significantly higher than those in other tissues(P<0.01),and the contents of glucagon in liver and heart were significantly higher than those in sandwich muscle,trapezius,latissimus dorsi,semitendinosus and external oblique abdominal(P<0.01),under different glycolytic potential(GP),the content of adrenaline in high GP group was significantly higher than that in low GP group(P<0.01).Among the genes,the m RNA expression of PFKM gene in heart was significantly higher than that in other tissues(P<0.01),and the m RNA expression of PYGM and PRKAG3 gene in sandwich muscle,trapezius,latissimus dorsi,gluteus medius,semitendinosus and external oblique abdominal was significantly higher than that in liver and heart(P<0.01).In correlation analysis,the expression of PFKM gene vary significantly positively correlated with the concentration of phosphofructokinase in trapezius(P<0.05),negatively correlated with free glucose and pyruvate kinase in liver(P<0.05),and negatively correlated with pyruvate kinase in heart(P<0.05);The expression of PYGM m RNA in trapezius was significantly positively correlated with the concentration of glucagon(P<0.05);The expression of GYS gene m RNA in external oblique abdominal was significantly positively correlated with the concentration of lactic acid(P<0.01),and negatively correlated with the concentration of glucagon in liver(P<0.01);The expression of glycolytic kinase in latissimus dorsi was significantly correlated with the expression of glycolytic kinase and glycolytic kinase(P<0.01).These results suggest that the glycolysis reaction is a complex biochemical metabolic process.There are obvious differences in glycolysis potential between different tissues,especially in liver and muscle tissues,and the related enzyme activities and hormone contents in liver are higher than those tissues.The expression of PFKM m RNA is highly expressed in heart tissue,which may participate in the glycolysis process of Yili horse mainly by affecting the related enzyme activity and hormone content in myocardial tissue.The expression of PYGM and PRKAG3 are highly expressed in skeletal muscle,which may mainly affect the glycolysis process of Yili horse by affecting the activity of related enzymes and the content of hormones in skeletal muscle.
