| In order to investigate the effects of diet on intestinal flora of chipmunk(Tamias sibiricus)under feeding conditions,as well as the relationship between dietary nutrients and changes in intestinal flora of chipmunk,a cage rearing experiment of chipmunk was conducted in March 2022 at the National Field Scientific Observation and Research Station of Maoermountain Forest Ecosystem,Heilongjiang Province.Wild chipmunks living in Maoer Mountain were captured and raised in this experiment.14 chipmunks were divided into field group,feed group,high fat group and high starch group with the nutrient content of diet as the variable.The fecal samples of chipmunks were collected regularly,and the intestinal flora of chipmunks in feces were sequenced by high-throughput sequencing after DNA extraction and PCR amplification.The main experimental results are as follows:1.16 S rRNA sequencing was used to analyze the intestinal flora of chipmunks.In this study,a total of 1845 OTUs were extracted from 60 chipmunks faecal samples.A total of 17 phyla,31 classes,72 orders,152 families and 335 genera were identified in OTU.The dominant bacteria were Firmicutes,Bacteroidetes and Proteobacteria,and the dominant bacteria at other classification levels were mostly from these dominant bacteria.2.When the food of chipmunks changed,the intestinal flora structure of chipmunks changed.At phylum level,there was no significant change in dominant bacteria in the diet group compared wit h the wild group,but the abundance proportion was different.Compared with the diet group,the abundance ratio of firmicutes in the high-energy group was decreased,and Bacteroidetes was the first dominant bacteria(t=-2.75,df =13,P = 0.0164).Compared with the diet group,actinomyces had the biggest change(t = 4.02,df = 13,P = 0.0015).3.Combined with the nutrient content in the food of chipmunks,the effects of nutrient composition on intestinal flora of chipmunks were analyzed.The content of crude protein in food samples was determined by Kjeldahl nitrogen determination method.The crude fat content in food samples was determined by Soxhlet extraction method.Fiber content in food samples was determ ined by filter bag fiber analysis.Combining the data of intestinal flora of chipmunks with the data of nutrient content in food,the results showed that the changes of intestinal flora of chipmunks were significantly correlated with nutrient composition.There was a significant correlation between actinomyces and crude fat content(r = 0.42,t = 3.08,df = 44,P = 0.0036).Crude fiber content was significantly correlated with Bacteroidetes(r = 0.50,t = 3.79,df = 44,P = 0.0005).Based on the above research results,it can be concluded that Firmicutes and Bacteroides are dominant bacteria in the intestinal flora structure of chipmurs,and dominant bacteria in other classification levels mostly come from these two dominant bacteria phyla.The composition,structure and diversity of intestinal flora of chipmunks changed significantly when the food was changed.There was significant correlation between dietary nutrient composi tion and intestinal flora of chipmunks. |
