| Fat mobilization caused by negative energy balance is a major factor in the high incidence of postpartum diseases in dairy cows.In Chinese medicine,abnormalities in lipid or lipid metabolism can be classified as "stagnant blood",while postpartum dairy cows are often suffering from Qi deficiency and blood stasis.The formula can be used to treat postpartum qi deficiency and blood stasis in dairy cows.The group found that different doses of the formula have the effect of reducing serum cholesterol levels in postpartum cows,suggesting that the formula has a certain regulatory effect on postpartum lipid mobilization in dairy cows.The effect of this formula in regulating and improving disorders of lipid metabolism is most likely one of the mechanisms of action in treating postpartum qi deficiency and blood stasis in dairy cows.Therefore,in this study,we evaluated the modulating effects of Guiqi Yimu oral liquid on lipid metabolism in postpartum cows in terms of liver lipid accumulation,blood metabolites and oxidized lipids.1.In order to investigate the intervention effect of Astragalus Yimou oral solution on fat accumulation in dairy cows,normal dairy cows were randomly divided into control group,fat accumulation group and Astragalus Yimou oral solution intervention group,and the lipid droplet content in hepatocytes of each group was observed by oil red staining,TG,ALT and AST content in hepatocytes of each group were measured by ELISA,and lipid metabolism in hepatocytes of each group was quantified by qPCR.The expression levels of PPARα,SREBP-lc,ChREBP,CPT1,CPT2 and MTP mRNA were quantified by qPCR.The results showed that the levels of lipid droplets,TG,ALT and AST in hepatocytes were significantly higher in the fat accumulation group compared with the control group(P<0.05),and 40g/L Guixi Yimu oral solution significantly reduced the levels of lipid droplets,TG,ALT and AST in hepatocytes(P<0.05),and the expression levels of PPARα,CPT1 and CPT 2 mRNA in hepatocytes were significantly lower in the fat accumulation group compared with the control group(P<0.05).The expression levels of PPARα,CPT1 and CPT 2 mRNA in the fat accumulation group were significantly lower than those in the control group(P<0.05),while the expression levels of ChREBP,SREBP-lc and MTP were significantly higher(P<0.05).40 g/L Astragalus membranaceus oral solution significantly reduced the expression levels of SREBP-lc,MTP and ChREBP mRNA(P<0.05).The results suggest that Astragalus membranaceus oral solution can alleviate the degree of hepatocyte injury and reduce intracellular lipid deposition.2.In order to elucidate the effect of Astragalus and Yiwu oral solution on the metabolic function of postpartum cows,15 Holstein cows with similar body condition and two to three litters of healthy cows awaiting parturition were randomly divided into control(n=7)and experimental groups(n=8),which were given drinking water and Astragalus and Yiwu oral solution at 500 mL/head immediately after parturition for 6 d.Blood was collected before morning feeding 7 d after parturition,and UPLC-MS-MS metabolomics and MetaboAnalyst 5 0 software were used to analyze the changes in plasma metabolic profiles and their related metabolic pathways in each group.The changes in plasma metabolic profiles and their associated metabolic pathways were analyzed in each group of cows using UPLC-MS/MS metabolomics and MetaboAnalyst 5.0 software.The results showed that the plasma metabolic profile was significantly altered in the test group compared with the control group,and 16 plasma metabolites were significantly altered(VIP≥ 1,P<0.05),of which 8 were significantly up-regulated,including methionine sulfoxide,creatine phosphate,glycerophosphorylcholine,cortisol,L-tryptophan,lysophosphatidylcholine(18:2),pentazocine and L-arginine;8 were significantly down-regulated,including 1aminocyclohexanoic acid,3-methylcrotonylglycine,L-asparagine,monoacylglycerol ester acyl(16:1),panthenyl mercaptoethylamine,octadecyl monoenamide,1-methylguanine and 3-N-methyl-L-histidine.The pathway enrichment results showed significant changes in glycerophospholipid metabolic pathway,arginine and proline metabolism,tryptophan metabolic pathway,and cortisol metabolism.The results suggested that the oral solution of Astragalus membranaceus had some regulatory effects on glycerophospholipid metabolism,arginine and proline metabolism,tryptophan metabolic pathway and cortisol metabolism in postpartum cows.3.To further analyze the modulating effect of Astragalus membranaceus Oral Liquid on lipid metabolism in postpartum cows,this experiment was conducted to detect the content of 26 oxidized lipids in the aforementioned plasma using UPLC-MS/MS lipidomics technique for quantitative analysis and pathway enrichment analysis.The results showed that the plasma oxidized lipid metabolic profile was significantly altered in the test group compared with the control group,and eight differential metabolites were screened(VIP≥1,P<0.05),of which five were significantly upregulated,including 12-HETE,11HETE,15-HETE,5-HETE,and 15-oxoETE;three were significantly downregulated,including 20-HETE,8,9-EET,and These differential metabolites were significantly enriched in arachidonic acid metabolism,inflammatory mediator regulation of TRP channels,PPAR signaling pathway,aldosterone synthesis and secretion,vascular smooth muscle contraction,and serotonergic synapses.The results suggest that Guiqi Yimu Oral Liquid may exert anti-inflammatory and antioxidant effects through modulating arachidonic acid metabolism and mediator regulation of TRP channels.Conclude:Astragalus Yunou Oral Liquid has a modulating effect on lipid metabolism in postpartum cows,can alleviate the degree of hepatocyte damage and reduce intracellular lipid deposition,and can regulate glycerophospholipid metabolism,arachidonic acid metabolism,and the mediator-regulated pathways of TRP channels in postpartum cows. |
