Effect Of Shenqi Polysaccharide Oral Liquid On Immune Functionin Immunosuppressive Chicken Induced With CY

In order to study the effect of Shenqi polysaccharide oral liquid on immune function in immunosuppressive chicken induced with CY.In the study,Pseudostellariae polysaccharide and Astragalus polysaccharide were obtained by extracting and purifying.Then,the purified Pseudostellariae polysaccharide and Astragalus polysaccharide mixed in proportion(called Shenqi polysaccharideoral liquid).Finally,Shenqi polysaccharide oral liquid was given to chicken by gavage to study its bioactive properties.The 180 SPF chicken were randomly divided into 6 groups,including control group,cyclophosphamide group(CY group),three polysaccharide oral liquid groups(low dose group,medium dose group,high-dose group)and positive control group(levamisole group).On the 10th day,the chicken in all groups except control group were intraperitoneally injected with CY80 mg/kg/d for 3 days,same volume physiological saline in control group.Then three polysaccharide oral liquid groups were administrated75,150,300 mg/kg/d for body weight Shenqi polysaccharide via gavage.The control group and the CY group were given physiological saline,and the positive control group was orally administered 20 mg/kg/d for levamisole,including 10-20 days,25-35 days.At the same time,the chicken in all groups were injected NDV-IBV-AIV(H9)vaccine at 14day and 28day.At 21 and 42 day,the organ index,the number of lymphocytes in blood,the titer of antibody in serum,the expression of sIg A,IL-4,IFN-? in the respiratory and intestine tract were determined.The changes of intestinal structure and the number of goblet cells were observed.The results were shown below:(1)The immune function index of each group showed:Compared with CY group,the spleen,thymus and bursa of the polysaccharide medium group were significantly higher(P<0.05);the number of lymphocytes in the low and high dose groups were significantly increased(P<0.05);the high dose group in Newcastle disease vaccine and Infectious bronchial vaccine and Avian influenza vaccine of antibody titers were significantly improved(P<0.05).The results indicated that Shenqi polysaccharide oral liquid could reverse the immune organ atrophy induced by cyclophosphamide immunosuppressed chicken,improve the immune function of ND and IB vaccine and immunefunction.(2)The expression of sIg A,IL-4 and IFN-y in the respiratory tract and intestinal tract of the chickens showed:The levels of sIg A,IL-4 and IFN-? in the mucosal mucosa and jejunum mucosa were higher than those in the CY group(P<0.05).It suggested that Shenqi polysaccharide oral liquid could repair the damage of immune function and improve the expression of related immune cytokines in chicken respiratory tract and intestinal tract.(3)The structure of the intestinal tract were observed:Compared with CY group,polysaccharide high dose group could significantly increase the intestinal villus height,intestinal V/C value and intestinal muscle layer thickness and the number of goblet cells(P<0.05).This means that Shenqi polysaccharide oral liquid not only could antagonize CY-induced damage of intestinal mucosal immune,but also could strengthen the intestinal mucosal immune function of chicken.
