The purpose of this experiment was to study the preventive effect of rosemary water extract on Eimeria tenella infection in yellow-feathered broilers,and its effects on growth performance,immune performance,anti-coccidiosis index,fecal oocyst value,antioxidant capacity and expression of inflammatory factors in yellow-feathered broilers.A total of 360one-day-old yellow-feathered broilers were randomly divided into 6 groups with 6 replications in each group and 10 chickens in each group.They were control group,negative control group,low,medium and high dose rosemary water extract adding group and positive control group.In the experiment,8×10~4 sporulated Eimeria tenella oocysts were fed orally except the control group at the age of 21 days.Two chickens in each group were randomly selected to collect serum,immune organs and cecum samples for 48 hours,96 hours,144 hours and 192hours after being infected,respectively.The preventive effect of rosemary water extract on Eimeria tenella in mitotic stage were determined.The results show that:The main results were as follows:The main results were as follows:(1)Although the addition of different doses of rosemary water extract had no significant effect on the growth performance,it could improve the feed-meat ratio of yellow-feathered broilers;compared with the positive control group,the anti-coccidial index of low-dose rosemary water extract group was similar to that of positive control group.the anti-coccidial index of middle and high dose rosemary water extract groups was slightly lower,but could achieve moderate anti-coccidial effect.(2)At 48 hours after being infected,compared with the negative control group,the immune organ index of positive control group and low,middle and high dose rosemary water extract groups were significantly improved(P<0.01),and 144 hours after being infected,compared with control group and negative control group,the bursa index of middle dose rosemary water extract group was significantly higher than that of control group and negative control group(p<0.05).The bursa index of middle dose rosemary water extract group was significantly higher than that of the control group and negative control group(p<0.05).At 192hours after being infected,the bursa index and spleen index in the low,middle and high dose rosemary water extract groups were significantly higher than those in the control group(p<0.05).(3)At 48 hours after being infected,the concentrations of T-AOC and MDA in positive control group and low,middle and high dose rosemary water extract groups were significantly lower than those in negative control group(p<0.05).96 hours after being infected,compared with the negative control group,the T-AOC concentration of positive control group and middle and high dose rosemary water extract group increased significantly,the SOD concentration of low and high dose rosemary water extract group decreased significantly,and the MDA concentration of positive control group and low and medium dose rosemary water extract group decreased significantly.Compared with the positive control group,the GSH-Px concentration of the remaining several being infected groups decreased significantly(p<0.05).At 144 h after being infected,compared with the negative control group,the concentrations of SOD and MDA in the positive control group and low,middle and high dose rosemary water extract groups were significantly lower,while the T-AOC concentration was significantly higher,while the GSH-Px concentration in the positive control group and high dose rosemary water extract group was significantly higher than that in the negative control group(p<0.05).192 h after being infected,compared with the negative control group,the SOD activity in the other being infected groups was significantly lower and the CAT activity was significantly higher than that in the negative control group(p<0.05).(4)Compared with the negative control group,the s Ig A content of positive control group and low dose rosemary water extract group increased significantly at 48 hours after being infected(P<0.01),and the LPS concentration of negative control group was significantly higher than that of other groups at 96 hours after being infected,and the s Ig A content of low,middle and high dose rosemary water extract groups was significantly lower than that of negative control group and positive control group(p<0.05).At 144 hours after being infected the concentration of DAO in the middle dose rosemary water extract group was significantly higher than that in the other groups,and the s Ig A content in the positive control group,low,middle and high dose rosemary water extract groups increased significantly compared with the negative control group(P<0.01).192 hours after being infected,the DAO concentration in the other groups was significantly lower than that in the negative control group,and the s Ig A content was significantly higher than that in the negative control group(P<0.01).(5)Compared with the Control group,the expression of IL-2,IL-6 and IL-8 in the experimental group increased significantly(p<0.05),the expression of IL-17 in the low dose rosemary water extract group was significantly higher than that in the other groups,and the expression of IL-17 in the middle dose rosemary water extract group was significantly lower than that in the other groups(p<0.05).Compared with the positive control group,the expression of TGF-βand Nrf-2 in the low,middle and high dose rosemary water extract groups decreased significantly(p<0.05),while the expression of TNF-αincreased significantly.y Compared with the negative control group,the expression of ZO-1 in the other groups decreased significantly(p<0.05).In summary,rosemary perfume extract can be used as a good anti-coccidial and growth-promoting plant additive in broiler diets. |
